| ObjectivePosttraumatic stress disorder(Posttraumatic stress,disorder,PTSD)is a serious disease caused by one or more catastrophic events,such as war,death threat,physical and psychological trauma,and violent crime.In recent years,with the increase of natural disasters,war and social violence and working pressure of life,the incidence of PTSD increased gradually,which seriously harms to the quality of the patient's life and even threatens the stability of the whole society.The treatment of PTSD is for psychotherapy and drug treatment,which SSRI antidepressant drug has become a first-line drug for post-traumatic stress disorder,but there are side effects in different degrees Clinical therapeutic effect is limited,even some drugs can cause drug addiction phenomenon.Therefore,it is urgent to clarify the pathogenesis of PTSD and the corresponding effective treatment measures.Our previous studies have proved that Wendan Decoction is effective to PTSD,and Ancient and modern studies have shown that Wendan Decoction is the best prescription for the prevention and treatment of PTSD,However,the "surprise is God out of the house," "God in the heart and in the kidney,then God does not shake.God do not pay the fine,is with "heart kidney disharmony,air floating situation is weaker,This provides the idea of "four" in the Jinding cardiac soup,on the basis of Wendan Decoction on Guizhi,Radix Paeoniae Alba,keel,oysters form Zhen Jing Wendan Decoction,mainly improve the PTSD panic symptoms.Therefore,the purpose of this study is to use Zhen Jing Wendan Decoction to improve the understanding of PTSD in Chinese medicine,and from the pathogenesis of qi stagnation,phlegm accumulation in the pathogenesis of this disease.By using the method of the whole experiment,to further clarify the effect of target Zhen Jing Wendan Decoction in the treatment of PTSD,and then from the ORX system to explore the neuro biological mechanisms of different levels,to provide new ideas for the pathogenesis of PTSD and clinical diagnosis and treatment etc.Methods1 First of all,this paper discusses the relationship between the PTSD and the five internal organs by combing the ancient literature with the theory of "panic",and focuses on the core pathogenesis of "Qi disorder,phlegm accumulation and depression".And the study focuses on improving the core symptoms of PTSD panic,Referring to the literature and modern research,Zhen Jing Wendan Decoction is based on Wendan Decoction,which modified cassia twig,white peony root,raw keel,raw oyster.2 The similarity of three batches of Zhen Zhen Wen Tang Decoction was determined by using the traditional Chinese medicine fingerprint.3 In this study,the SPS model was used as stressor.Through open field test,stiff test,Elisa testand TEM,the PTSD model was evaluated from two levels:physiological state behavior change.On the basis of this,we discuss the regulating mechanism of Zhen Jing Wendan Decoction.4 From the cerebrospinal fluid to gene level,using Elisa and real timePCR technique,to explore the mechanism of PTSD model and Zhen Jing Wendan Decoction for the ORX mediated synaptic plasticity.Results1 the chromatogram of the samples in the three batches of the decoction is basically the same.It can be used for experimental research.2 through the open field test and the stiff experiment,the PTSD model of SPS replication was evaluated from the behavioral level,and on the basis of the above,the regulation effect of the decoction was analyzed.From the perspective of behavior science,the results of open field test for the rat model of 7day by SPS,the number of rearing,total movement distance,the number of crossing line,the central cell residence time and the maximum movement distance,the two groups had significant difference compared with the normal group(P<0.05).Compared with the model group,the total motion distance,the total movement distance,the number of wearing,the residence time and the maximum moving distance were improved,and the difference was statistically significant(P<0.05).Among them,the difference of the total movement distance and the number of wearing grid was significantly different from that of the model group(P<0.01).Compared with the model group,the western medicine group had a significant improvement in the number of times,the total distance of movement,the number of wearing,the residence time of the central column and the maximum moving distance.There were no significant differences between the groups in the number of times,the total distance of movement,the number of wearing,the length of stay and the maximum movement distance.The results of experiment for the rats froze after SPS7d,the rats in the normal group freezing time compared with the model group,and there was significant difference(P<0.01),each treatment group and model group had significant difference(P<0.01).the anticonvulsant Wendan Decoction and paroxetine group compared with the normal group,no statistical learn the difference,Wendan Decoction was significantly different compared with normal group(P<0.01).3 the model of PTSD replication by SPS was evaluated from the aspects of stress hormones and the changes of brain function,and on the basis of the above,the regulation of was also studied.The results of serum corticosterone Elisa after SPS modeling 7d,the rat serum corticosterone concentration model group was significantly higher than the normal group,and there was a significant difference(P<0.01).Compared with the model group,there was significant difference between the two groups(P<0.05),but there was significant difference between the two groups(P<0.01).Among them,the difference between the Zhen Jing Wendan Decoction,the paroxetine group and the model group was more obvious(P<0.01),and there were also differences between Wendan Decoction,Wendan Decoction and Zhen Jing Wendan Decoction(P<0.05).The results of serum neuron specific enolase(NSE)Elisa after SPS,the serum concentration of NSE in the model group was significantly higher than that in the normal group,and there was significant difference(P<0.01).Compared with the model group,there was statistical significance(P<0.01).Compared with the normal group,there was no difference between the Wendan Decoction group and the normal group(P<0.01).The results of transmission electron microscopy showed that the neurons in the prefrontal cortex of the normal group had a complete and clear neuron shape and nuclear membrane,and the chromatin distributed evenly.In the model group,the nuclei of neurons in the prefrontal cortex were deformed,and the morphological changes of the nuclear membrane with multiple folds and depression.Chromatin is condensed to the nuclear membrane and the distribution is very uneven.Each group of neurons was Oand the majority of condensation on the nuclear membrane,mostly crescent shaped change,nuclear membrane folds or subsidence,even deformed nuclei.In the normal group,the hippocampal neurons were more complete and clear,the chromatin distributed evenly and the nuclear membrane was clear.The neurons and the model group were improved to varying degrees.4 the results of OrexinA and OrexinB in cerebrospinal fluid(CSF)of rats were detected by Elisa test.The contents of OrexinA and OrexinB in cerebrospinal fluid of model rats were higher than those in normal group(P<0.01).There was a difference between the two groups(P<0.05).Among them,in the aspect of cerebrospinal fluid Orexin A,there was a significant difference between the group of Wendan Decoction and Wendan Decoction and paroxetine group(P<0.05).Compared with the model group,the difference of Orexin Aand B in cerebrospinal fluid(CSF)was significantly higher than that of model group(P<0.01).The results of real timePCR showed that the OX1R and OX2R of PTSD rats in the model group HIP,PFC and AMY were significantly decreased compared with the normal group,and there was significant difference(P<0.05).The treatment group increased in HIP,PFC,AMY and OX1R,the expression of OX2R,and compared with the model group improved in varying degrees in the prefrontal cortex,convulsion of Wendan Decoction group and the control group was similar to paroxetine group in up regulation of OX1R,there are differences with the model group(P<0.05),and no difference with the normal group.In the amygdala,anticonvulsant effect of Wendan Decoction group increased OX1R have significant differences with the model group(P<0.05),and the effect of Western medicine was similar to the control group,no difference compared with the normal group,Wendan Decoction group compared with the normal group there are still differences(P<0.05).Zhen Jing Wendan Decoction group raised in OX2R compared with the model group had significant difference,the effect is better than that of paroxetine group and Wendan Decoction group.Conclusions1 From the two levels of behavioral and physiological state,it is proved that the single extended stress(SPS)method can be used to replicate the panic and anxiety of PTSD rats.The physiological changes in Zhen Jing Wendan Decoction regulating PTSD induced down-regulation,treatment effect is better than that of Wendan Decoction group,the serum NSE concentration of serum corticosterone.The description of Zhen Jing Wendan Decoction in improving the physiological state of PTSD induced superior Wendan decoction.2 The experimental results show that PTSD can affect the content of OrexinA and OrexinB in cerebrospinal fluid of rats.The results of immunohistochemistry and real timePCR showed that the changes of OX1R and OX2R receptor and the level of gene in the cortex of PTSD like rats were basically the same.It is suggested that the activation of ORX neurons in the brain,due to the intervention of a single prolonged stress method,can cause stress and anxiety.During the process of PTSD stress,the increase of OrexinA and OrexinB in CSF and the decrease of OX1R and OX2R receptor content.This leads to the high activation of ORX neurons in different brain regions,which leads to the generation of PTSD stress response,such as anxiety and fear.3 Zhen Jing Wendan Decoction and Wendan Decoction can improve the symptoms of PTSD and anxiety,which shows that the traditional Chinese medicine prescription of regulating qi and resolving phlegm can improve the symptoms of fear and anxiety of PTSD.According to the above results,Zhen Jing Wendan Decoction improving PTSD is better than Wendan Decoction,It shows that the traditional Chinese medicine therapy,which is based on Regulating Qi and resolving phlegm,can effectively improve the anxiety and fear of PTSD effectively.Its role may be related to the regulation of OrexinA,OrexinB,OX1R and OX2R receptor in ORX system. |
