Purpose:TO observe Shiwei Wendan Decoction’s effect on the central nervous system,and influence on the animal models of schizophrenia behavior.Methods:In experiment 1,the sleep latency and sleep time in mice was determined by intraperitoneal injection of sodium pentobarbital(40 mg/kg)to observe Shiwei WendanDecoction’s influence on the sleep of mice caused by threshold dose of pentobarbital.In experiment 2,the convulsive incubation period and survival time in mice was determined by intraperitoneal injection of e four nitrogen method(110 mg/kg)to observe Shiwei WendanDecoction’s influence on the convulsions in mice and discuss Shiwei WendanDecoction’s influence on the central nervous system. In experiment 3, with subcutaneous injections of palmer jilin(75mg/kg)first,and then intraperitoneal injection of 5- hydroxy tryptophan(20mg/kg),we observed the glancing frequency of model mice in five minutes.In experiment 4,we established model mice of isolation solitary aggression to observe Shiwei WendanDecoction’s influence on the attack behavior, in order to investigate Shiwei WendanDecoction’s influence on behavior related to animal models of schizophrenia.Results:Experiment 1show that, compared with the blank group, Shiwei Wendan Decoction can significantly shorten the sleep incubation period of mice, P<0.05, estazolam group had shorten the sleep latency in mice extremely, P<0.01. meanwhile,estazolam group was better than Shiwei Wendan Decoction. Compared with the blank group, the sleep durations of Shiwei Wendan Decoction and estazolam group were both extended obviously, P<0.01. meanwhile, estazolam group was better than Shiwei Wendan Decoction.Experiment 2 shows that,compared with the blank group, the convulsions latency and survival time of phenytoin sodium group and Shiwei Wendan Decoction group were both significantly prolonged, P < 0.01. Meanwhile,phenytoin sodium group was better than Shi wei Wendan Decoction group.Experiment 3 show that,compared with the blank group, the number of Head Thrust Test of the haloperidol group and the Shiwei Wendan Decoction group were significantly r educed after 3days or 10 days respectively,P<0.01.Compared with Shiwei Wendan Decoction group, the number of Head Thrust Test of Haloperidol group is less after three days dosi ng, P<0.05,the number is significantly reduced for ten days, P<0.01.3days or 10 days after t he treatment compared with itself before dosing respectively, the number of Head Thrust T est of Shiwei Wendan Decoction group and haloperidol group were significantly decreased,P<0.01. The number of Head Thrust Test of Shiwei Wendan Decoction group dosing 10 days is less than 3 days, P < 0.05. Haloperidol group who got 10 days dosed, whose the number of Head Thrust is significantly reduced than 3 days,P<0.01.Experiment 4 shows that the attack score had no significant difference in each group before the medicine. Compared with the blank group after the treatment, attack scores com e from Shiwei Wendan Decoction group and buspirone group were reduced, P<0.01.Buspir one is compared with Shiwei Wendan Decoction group,the attack scores of Buspirone gro upwere reduced, P<0.01. Each group is dosed before and after in comparison shows there are differences in the above indexes, P<0.05.Conclusion: Shiwei Wendan Decoction has certain inhibitory effect on the central ne rvous system, can reduce the schizophrenia head count and aggression behavior experiment model mice.
