The Expression Of Allergic Eosinophil-associated Factor And The Intervention Mechanism Of Allergic Health

Objective:Looking for the difference between allergic constitution and balanced constitution,and finding out the effective specific molecular markers of allergic constitution.Allergic rhinitis Subjects were treated for traditional Chinese formula "Guominkang" by conditioning allergic constitution.The researchers observed the clinical effects and explored the target of the TCM formula "Guo Min Kang" conditioning allergic constitution to cure allergic disease.In addition,the last purpose of this study is to search the relationship between the mechanism and the regulation of eosinophil function.The last but not the least,to investigate the micro-mechanism of allergic constitution may provide better guidance for the clinical application of the concept of allergic constitution.Methods:Basing on the Classification and Determination of Constitution in TCM and the allergen test,we included in the 52 subjects by post recruitment poster and collection the patients who come to Chinese medicine clinic for treatment.The ratio of male to female was 1:1,and their age all between 18 to 40 years old.The subjects were required to sign informed consent.There were 32 in allergic constitution group as experimental group,and20 ones in balanced constitution group as control group.Subjects of allergic constitution group were received 3 months’ medical intervention,and there was no any intervention measure in the control group.Convenient for experiment description,the allergic constitution group after treatment was named as allergic group after intervention.In the following experiments,the samples of each group were detected by protein chip,QGP and ELISA method.Blood samples of subjects in the three groups were all collected separately on an empty stomach in the morning,collect 20ml of blood on empty stomach in the early morning.The sample is centrifuged to obtain the serum.The levels of IL-5,GM-CSF,IL-3,eotaxin,RANTES,IL-4,IL-13,and IL-9 protein were detected by protein chip technology.The levels of IL-25,IL-33,TLSP and ECP protein were detected by ELISA Kit.5ml venous blood was extracted from the patients with EDTA anticoagulant tube,Take 36ul whole blood for pretreatment according to the manual of QuantiGene plex kit.The levels of IL-5,eotaxin and CCR3 mRNA were detected by QuantiGene Plex method.Data processing were analysed with spss22.0 software.The independent t test method was used to evaluate the difference between balanced constitution and allergic constitution.The paired t test was used to evaluate the changes of the indexes before and after the intervention of allergic constitution.the data were expressed as x±s,P<0.05 means statistical difference.Results:1.Used the TCM formula "Guo Min Kang" conditioning allergic constitution to cure allergic disease,after three month,the scores of TNSS,TNNSS,RQLQ were decreased.The score of main evaluation index(TNSS),including nasal congestion,runny nose,nasal itching and sneezing.The four nasal symptoms improved significantly from fourth to eighth weeks in the treatment,and for the next 4 weeks after treatment,the score continued to decline.In a return visit after the end of the experiment,by comparison,it can be found that allergic symptoms and living conditions of the subjects in the second allergic seasons after taking TCM formula were significantly improved than the end of the test.It is worth mentioning that,some subjects were not recurrence at all.2.After the intervention,there was no significant decrease both in tIgE and sIgE serum levels in subjects of allergic constitution.3.The difference factors between allergic constitution and balanced constitution:Comparison between the two groups,the levels of IL-5,GM-CSF,eotaxin,RANTES,IL-4,IL-13,IL-9,IL-25,TLSP,IL-33,ECP protein and the levels of IL-5,eotaxin,CCR3 mRNA in the subjects of allergic constitution were higher than those in balanced constitution,and the difference was statistically significant(P<0.05).The levels of IL-3 protein in the subjects of allergic constitution were higher than those in balanced constitution,but the difference has no statistical significance(P>0.05).4.Used the TCM formula "Guo Min Kang" conditioning allergic constitution to cure allergic disease,after three month,the levels of IL-5,eotaxin,RANTES,IL-4,IL-13,IL-25,TLSP,ECP protein and IL-5,eotaxin,CCR3 mRNA in the subjects of allergic constitution were lower than those before the intervention,and the difference was statistically significant(P<0.05).The levels of GM-CSF,IL-9,IL-33 protein in the subjects of allergic constitution were lower than those before the intervention,but the difference has no statistical significance(P>0.05).Conclusion:1.TCM formula "Guo Min Kang" conditioning allergic constitution to cure allergic disease can effectively improve the clinical symptoms and quality of life,and this improvement is sustained.2.IL-5,GM-CSF,eotaxin,RANTES,IL-4,IL-13,IL-9,IL-25,TLSP,IL-33,ECP,CCR3 is the factor differences between allergic constitution and balanced constitution.3.The mechanism of TCM formula "Guo min kang" conditioning allergies involved in the regulation of eosinophil related functions.IL-5,eotaxin,RANTES,IL-4,IL-13,IL-25,TLSP,ECP,CCR3 may be the target of "Guo Min Kang" conditioning allergic constitution.
