Study On The Molecular Expression Of Allergic IgE Antibody Class Conversion And The Intervention Mechanism Of Allergic Health

OBJECTIVE:To define the differences in the expression of IgE antibody CSR related molecular in the allergic constitution and peace constitution by molecular biology technique,to find the molecular biology markers of allergic specificity.To explore the microscopic mechanism of allergic constitution formation;to observe the improvement of allergy-related diseases and quality of life of allergy groups after the optimization and intervention of Guominkang capsule for allergic constitution;to definite effect target of the IgE antibody CSR regulate process in allergic constitution by Guominkang capsule.Methods:According to the classification and determination of Chinese medicine constitution,allergen test and allergic rhinitis symptom score table,52 people,including allergic constitution in 32 cases,peace constitution in 20 cases,excluding those who have multiple constitution,clear diagnosis of Western medicine and are in medication period and pregnant or lactating women,in which the male to female ratio is 1:l,age were between 18 to 40 years old.The source of the subjects was divided into two parts,one was from patients with allergic rhinitis of the Chinese medicine outpatient department in Beijing University of Traditional Chinese Medicine Guanyi Chinese medical center,Beijing Shengyi medical center,Beijing Hongyi medical center;the other was from the student volunteers of major colleges in Beijing recruited by the"Recruitment of allergic and peace constitution tester" posters.Taking the fasting venous blood from subjects above in the morning for 10ml,preprocess the whole blood and separation of serum based on QuantiGene plex kit instructions.The expression differences of IL-4,IL-13,CD40,BAFF,APRIL and IFN-? in serum can be detected by protein chip technique.The expression level of AID and Id2 mRNA can be detected by QGP method.After three months of intervention by Guominkang capsule,take the peripheral blood from allergic patients and repeat the tests above.The expression levels of serum total IgE and sIgE before and after treatment can be detected by immunoCAP.Data acquired can be analyzed by spss20 software and paired t test,with x ± s,P<0.05 stand for was statistically significant,P>0.05 stand for was not statistically significant.Observing the changes in TNSS,TNNSS and RQLQ changes of the Patients with allergic rhinitis respectively before the Guominkang capsule intervention,4 weeks after intervention,8 weeks after intervention,and 12 weeks after intervention and 1 year after intervention.Datas were analyzed by spss 20 software,two samples were compared,the difference was normal distribution,using paired t test.The difference is normal distribution,using wilcoxon rank sum test.Metrological data were expressed as x ± s,P<0.05 was statistically significant.Outcome:1.The levels of IL-4,IL-13,CD40 in the serum of the allergic constitution group were significantly higher than those of the control group(P<0.05).APRIL,BAFF protein concentration was higher than that of the peace constitution group,the difference was not statistically significant(P>0.05).The expression of AID mRNA in the allergic constitution group was significantly higher than that in the peace constitution group(P<0.05).The expression level of Id2 mRNA and IFN-? was lower than that of the control group,the difference was statistically significant(P<0.05).2.The levels of IL-4,IL-13 in the serum of the allergic constitution group after the Guominkang capsule intervention was lower than those before the intervention,the difference was statistically significant(P<0.05).CD40,BAFF and APRIL protein concentrations were lower than those before intervention,the difference was not statistically significant(P>0.05).The expression level of AID mRNA was lower than that before the intervention in allergic constitution group,the difference was statistically significant(P>0.05).The expression level of IFN-? was higher than that before the intervention,the difference was statistically significant(P<0.05);The expression level of ID2 mRNA was higher than that before the intervention,the difference was not statistically significant(P>0.05)3.Compared with pre-intervention,the TNSS,TNNSS and RQLQ scoring of allergic rhinitis patients were decreased after the Guominkang capsule intervention for 4 weeks,8 weeks,12 weeks and 1 year,the differences were statistically significant(P<0.05).Conclusion:1.IL-4,IL-13 and IFN-y are the differentiating factors of peace and allergic constitution,and can be used as the effect target of the allergic constitution process by Guominkang capsule;The differences between peace and allergic constitution is related to the regulation process to AID induction by the synergistic relationship between IL-4 and CD40 during the antibody CSR;The research suggested that the difference in IgE antibody CSR between allergic and peace constitution may not involve APRILand BAFF as an alternative route;The lack of Id2 plays an important role in allergic disease.2.In the process of antibody CSR,there were many transcription factor-binding sites involved,and Guominkang capsule may directly act on NF-?B,STAT6,E2A and other transcription factor binding sites or protein kinases in the process of antibody CSR,and regulate allergic constitution,which remains to be further studied in the future.3.The level of IgE expression in serum may be related to the affinity of IgE produced by antibody CSR.Low affinity IgE and high affinity IgE can lead to beneficial competition to prevent the occurrence of allergic reactions,and after the intervention,IgE decreased insignificantly,but allergic symptoms were improved,which may be the result of low affinity IgE effect.It remains to be further validated by later experiments.4.Guominkang capsule can regulate the allergic constitution in the respect of "Fu Zheng Qu Xie",which also can significantly improve the symptoms of patients with allergic rhinitis.However,constitution is a long-term stable state of existence,the change can be very slow,and so the latter part of the experiment need to further increase the medication time,guidance for the patient's diet and living can further improve their constitution.
