Objective:In this study, FD mice is considered as a research object. ICC and SMC which effects gastrointestinal motility are regarded as a target. We will discuss the effect and mechanism of acupuncture treatment of functional dyspepsia from ghrelin and SCF/ c-kit signaling pathway that is the key point influencing ICC and SMC.Methods:60 SPF c57bl mice were randomly divided into 5 groups:control group, model group, acupuncture group, ACK2 model group, ACK2 acupuncture group. In addition to the control group, the other four groups of mice were modeled in accordance with starvation and overeating combined with L-arginine (L-arginine, L-Arg).At the fifth day of modeling,the ACK2 model group and ACK2 acupuncture group were injected with c-kit neutralizing antibodies (ACK2),50?g/d for 10 days, and the rest were injected with the same dose of saline. After modeling, all mice no food but drink should be given 24h,and each group were randomly selected 6 mice, then these mice were given by intragastric administration with a semi-solid paste 0.5g/only.15 minutes later, mice were sacrificed by taking off the neck, then open the abdominal cavity to take gastric tissue, detect gastric emptying rate, and observe the changes of gastric mucosa group of pathological tissue in the light microscope. The acupuncture group and ACK2 acupuncture were given a treatment time of 7 days. After acupuncturing treatment, no food but drink should be given 24h, using the method of intragastric administration with a semi-solid paste 0.5g/only to detect gastric emptying, radioimmunoassay to measure Ghrelin in serum, antral tissue and CCK, Western blotting (Western Blot) to test SCF in gastric antrum and c-kit protein, QPCR to discover the expression of SCF and c-kit mRNA in gastric antrum, and immunofluorescence to detect ICC and SMC optical density.Results:1. Comparison of food intake and body weight of mice in each group:Compared with the control group, model group mice's food intake and body weight decreased significantly (P<0.05).Compared with model group, acupuncture group mice's food intake and body weight increased significantly (P<0.05).2. Comparison of gastric emptying in each group of mice:Compared with the control group, model group were significantly increased gastric residual rate, the difference was statistically significant (P<0.05).Compared with the model group, the gastric residual rate of acupuncture group were significantly lower, the difference was statistically significant (P<0.05).3. Comparison of serum and gastric antrum's Ghrelin in each group:Compared with the control group, model group serum and gastric antrum's Ghrelin content are both decreased, the difference was statistically significant (P<0.05). Compared with the model group, Serum and gastric antrum tissue's Ghrelin in acupuncture group were significantly increased, the difference was statistically significant (P<0.05).4. Comparison of serum and gastric antrum's CCK in each group:Compared with the control group, serum and gastric antrum's CCK content were all increased, the difference was statistically significant (P<0.05).Compared with the model group, serumand antrum tissue's CCK in acupuncture group were significantly lower, the difference was statistically significant (P<0.05).5. Comparison of SCF and c-kit protein in gastric antrum:Compared with the control group, model group's SCF and c-kit protein content in gastric antrum tissue was significantly lower, the difference was statistically significant (P <0.05).Compared with the model group, acupuncture group's SCF and c-kit protein levels in gastric antrum were significantly increased, the difference was statistically significant (P<0.05), ACK2 model group's content of c-kit in gastric antrum was significantly lower, the difference was statistically significant (P<0.05).6. Comparison of mRNA's expression in gastric antrum's c-kit in each group:Compared with the control group,model group's expression of mRNA in SCF and c-kit in gastric antrum was lower, and the difference was statistically significant (P<0.05).Compared with the model group,acupuncture group's expression of mRNA in SCF and c-kit in gastric antrum was rose,ACK2 model group's expression of mRNA in SCF and c-kit in gastric antrum was significantly increased, the difference was statistically significant (P< 0.05).7. Comparison of optical density value of c-kit receptor in ICC:Compared with the control group,the optical density value of c-kit receptor in ICC was reduced,which were in gastric antrum in model group, the difference was statistically significant (P <0.05). Compared with the acupuncture group,the optical density of c-kit receptor in ICC was decreased,the difference was statistically significant (P<0.05).8. Comparison of VAchT receptor's optical density in SMC:Compared with the control group,the optical density of VAchT receptor in SMC was decreased,the difference was statistically significant (P<0.05).Compared with the model group,the optical density of VAchT receptor in SMC was increased,the difference was statistically significant (P<0.05).Compared with the acupuncture group,the optical density of VAchT receptor in SMC was decreased,the difference was statistically significant (P<0.05).Conclusions: 1. Acupuncture can effectively increase food intake and body weight,accelerate gastric emptying movementfurther.improve gastrointestinal motility.2. Acupuncture can improve Ghrelin and CCK's content, so as to improve gastrointestinal motility.3. Acupuncture can increase the content of SCF and c-kit protein and the expression of mRNA to improve SCF/c-kit signaling pathway, thus it can improve the function of ICC to adjust the gastric motility.4. Acupuncture can increase the optical density value of the receptors in c-kit and VAchT which are in ICC and SMC, and promote the recovery of ICC and SMC net structure and function,further improve gastrointestinal motility.5. Acupuncture regulate brain-gut peptide content by SCF/c-kit signaling pathway, so as to change the function of ICC and SMC.
