ObjectiveTo established a rat model of early AS and at the same time the Shenqi Jiyu Tang is used in the rat model in order to assess synthetically the effect of medicated decoction in prevention of AS from the general situation of rats, such as changes of blood lipid metabolism, inflammatory response, organ pathological changes etc. Try to explore the scientific nature and rationality of medicated diet decoction through the study of components of Shenqi decoction. And provide a theoretical basis for clinical prevention of AS by using the Shenqi Jiyu Tang.MethodsSeventy-two male Wistar rats were randomly divided into six groups:blank group, model group, positive control group (referred to as positive group), Shenqi Jiyu Tang group (referred to as S JT group), Danshen Huangqi Tang seperated prescription group (referred to as ST group)and Jiyu tang prescription group (referred to as JT group). There were 12 rats in each group. After 7 days adaptive feeding, except the blank group, other 5 groups were fed with high-fat diet combined with intraperitoneal injection of VitD3 to establish a rat model of early AS. At the same time, the model was intervened:SJT group were fed with the Shenqi Jiyu Tang, the ST group were fed with Danshen Huangqi Tang, the JT group were fed with Jiyu Tang, the Positive group group were fed with atorvastatin calcium solution, the blank group and the model group were fed with normal saline. Each group was fed with once a day, the dose was 1ml/100g/d and the intervention time was 12 weeks. Rats were observed daily and the body mass was measured. The rats were sacrificed at twelfth week after blood collection. Determination of serum lipids levels, including the level of TC, TG, HDL-C and LDL-C, and AI and determination of serum hs-CR level. In order to observe the pathological changes of the aorta, to calculate the liver coefficient and observe the hepatic pathological changes.ResultsTo the end of 12 week, a total of 64 rats were completed the experiment,8 rats were died. The experimental results are as follows:1. General situation and the change of body mass(1) General situation:in the process of intervention, the blank group rats were normal. Rats in the model group appeared withered hair color dull, unresponsive, weakness, fatigue, and other phenomenon. Compared with the model group, the intervention group had different degrees of improvement, the SJT group and JTgroup were the lightest.(2) Body wight:The SJT group and JT group were higher than that of the other Intervention groups (P< 0.05). Before the intervention and after the intervention of 4th,8th and 12th week, the variance analysis of repeated measures analysis of 4 time points was showed:In the 6 groups, there were significant in the group effect, time effect and the interaction effect (P< 0.05). In the process of intervention, the whole process of blank group showed an increasing trend;Exdept the blank group, the rest of 5 groups showed a slow downward trend.After 4 to 12 weeks, the SJT group and JT group showed a stable trend of body mass. And the quality of the rest of the intervention groups showed a downward trend.2. The four level of blood lipids and arterial stiffness index (AI) changes(1) Serum TC level:The other 5 groups were higher than the blank group (P< 0.05); the positive group, the SJT group and ST group were lower than that of model group (P< 0.05),there was no significant difference (P> 0.05) between model group and JT group; the SJT group and ST were higher than that of positive group (P< 0.05); the SJT group and ST group had no statistical significance (P>0.05).(2) Serum TG level:The other 5 groups were higher than the blank group (P< 0.05); the SJT group, ST group and JT group were lower than the model group (P< 0.05),there was no significant difference between the SJT group and the positive group (P> 0.05); the SJT group was higher than that of positive group (P< 0.05); the Shenqi tang group and JT group was higher than that of the SJT group (P< 0.05).(3) Serum HDL-C level:The other 5 groups were lower than the blank group (P< 0.05); the positive group, SJT group and ST group were higher than the model group (P< 0.05),there was no significant difference between the JT group and the model group (P> 0.05); there was no significant difference between the SJT group and the JT group;the ST group and the SJT group had no statistical significance (P> 0.05).(4) Serum LDL-C level:The other 5 groups were higher than the blank group (P< 0.05);the Positive group, the SJT group and the ST group was lower than model group (P< 0.05),there was no significant difference between the JT group and the model group (P> 0.05); the SJT group and the SJ group were higher than the positive group;ST group and the SJT group had no statistical significance (P>0.05).(5) AI:The model group and the JT group were higher than the blank group (P< 0.05); the Positive group, the SJT and the ST group were lower than model group (P< 0.05),there was no significant difference (P>0.05) between the JT group and the model group;there was no significant difference in AI between the SJT group and the ST group(P>0.05);the SJT group and the ST group had no statistical significance (P>0.05).3. Changes of serum hs-CRP levelsThe other 5 groups were higher than the blank group (P< 0.05); the Positive group, SJT group and ST group was lower than the model group (P< 0.05);there was no significant difference between the JT group and the model group(P>0.05); there was no significant difference comparing the SJT group and the ST group with the positive group (P>0.05); Compared with the SJT group, there was no significant difference in the ST group (P>0.05).4.Pathological changes of aortaIn the blank group, the aortic tissue was normal. The structure of each layer of the model group was not clear, VSMC in the membrane disorder, loose, partial loss of dothelial cells. There are a few inflammatory cell infiltration in the inner foam cells. Compared with the model group, the pathological changes of the aorta in the other intervention groups had different degrees of improvement. The main performance of the aortic structure is clear, in the membrane VSMC arrangement is loose, endothelial cells were more complete, almost no foam cells, etc. The positive group of the aortic pathological improved significantly, aortic pathological morphology in the SJT soup group improved obviously.5. Liver pathological changes and liver coefficient(1) Liver pathological changes:The liver morphology observation showed that there was no abnormality in the blank group. The hepatic cells of model group were obviously swollen under light microscope, and the nucleus was squeezed to one side, appear like bullous steatosis, inflammatory cell infiltration liver sinus congestion. The rest of the intervention group had different degrees of improvement in the liver, liver pathological morphology improved significantly in the positive group and S JT group.(2) Change of liver coefficient:The other 5 groups was higher than that of the control group (P< 0.05). The positive group and SJT group was lower than that of model group (P < 0.05), there was no significant difference between the ST group and JT group with the model group (P>0.05).ConclusionsShenqi Jiyu Tang can prevent the rat of early AS, including improving the general condition and body weight,regulating the blood lipid abnormality,reducing the hs-CPR, control the aortic lesion and hepatic steatosis in AS rats. Using mixed SJT to prevent the early AS in rats is better than the disassembled prescriptions groups. This study shows that our diet is scientific and reasonable. Therefore the Shenqi Jiyu Tang is a reasonable method to prevent AS. It is worthy of further research and clinical promotion.
