Objective: In order to analyze the correlation between C-erb B-2, the Ki67, P53, CA15-3 and AKP expression and bone metastases in breast cancer, we detected C-erb B-2, the Ki67, P53 on m RNA and protein expression levels in patients and CA15-3, AKP content in their serum. This may be valuable for the exploration of clinical value of these markers in clinical diagnosis of bone metastases in breast cancer. At the same time, we researched cellular immune function in breast cancer patients with bone metastases and without metastases group. we analyzed the expression level total of T cells, T helper cells, cytotoxic T cells, B lymphocyte, CD4 / CD8 and NK cells in breast cancer patients with bone metastases in order to provide reference for clinical treatment and prognosis of patients with breast cancer.Methods: Screening of breast cancer patients visited cancer center of Zaozhuang municipal hospital from January 2010 to December 2014, we identified a total of 49 patients with bone metastases of breast cancer as the experimental group, a total of 29 patients with breast cancer without metastases as control group. The m RNA expression level in the breast cancer tissue were detected with real-time quantitative PCR. Immunohistochemical method was used to detect C-erb B-2, Ki67 and P53 expression level in patients. The expression of these factors were calculated with grey value and positive rate of accumulative integral method. Serum CA15-3 content was detected with chemiluminescence method. Serum alkaline phosphatase level was detected by rate of biochemical method. Using flow cytometry instrument to detect cellular immune function in patients, such as, the expression of total T cells, T helper cells and cytotoxic T cells, B lymphocyte, CD4/CD8 and NK cell.Results: The expression of C-erb B-2, Ki67 and P53 in breast cancer with bone metastasis were higer than these of breast cancer without metastases. Compared with the non metastases group, the m RNA expression of C-erb B-2, Ki67 and P53 in breast cancer with bone metastasis were 9.05?29.04?33.44. The protein expression of C-erb B-2, Ki67 and P53 in breast cancer with bone metastasis in bone metastasis were 1.59, 2.47 and 2.51, while the control group were 0.41,1.07, and 1.03, There was significant difference between bone metastases group and the control group(C-erb B-2, P<0.001;Ki67, P<0.01;P53, P<0.001).Serum content of CA15-3 and AKP in bone metastases group were 25.81 and 123.57.While the control group were 12.17 and 85.59. There was significant difference between bone metastases group and the control group(CA15-3, P<0.001;AKP, P<0.01).Positive rate of C-erb B-2, Ki67, P53, CA15-3, AKP and joint inspection in breast cancer with bone metastases group were 79.59%?77.56%?93.87%?26.53%?44.9%. The Positive rate of these factors and joint inspection in control group were 41.38%?58.62%?55.17%?6.9%?27.59%. There was statistical significance between bone metastases group and the control group. The sensitivity of these factors in breast cancer were 79.59%?77.56%?93.87%?26.53%?44.9%. The specifity of these factors were 58.62%?41.38%?41.38%?93.1%?72.4%.Furthermore, we analyzed the joint inspection of these factors. When these factors were combined with one and another, the combination of the highest specificity was AKP and CA15-3, with specificity of 86.21%. The combination of the highest sensitivity was C–erb B-2 and P53, sensitivity of 97.96%. The sensitivity of the detection increases with more factors detected, while the specificity of the detection was reduced.According to the cellular immune function results, T cells in breast cancer bone metastases group was lower than that of breast cancer without metastases group(P < 0.01). T helper cells in breast cancer bone metastases group was significant lower than that of breast cancer without metastases group(P < 0.001). Cytotoxic T cells significant higher than breast cancer without metastasis group(P < 0.05). CD4 +/CD8 + in breast cancer bone metastases group was significantly lower than that of breast cancer without metastases group(P < 0.001).NK cells and B cells in breast cancer with bone metastases was lower than breast cancer without metastases group, but there was no significant difference.Conclusion: The expression of C-erb B-2, the Ki67, P53 expression and serum CA15-3, AKP were significantly increased in patients with bone metastases. There may be certain relevance between the expression of C-erb B-2, Ki67, P53, CA15-3, AKP and bone metastases. The combination of these markers has clinical value for the prediction and diagnosis of bone metastases of breast cancerWhen the combination of these each two markers, C-erb B-2?Ki67?P53?CA15-3 and AKP, the joint inspection of AKP and CA15-3 has the highest specificity. While the joint inspection of C-erb B2 and P53 has the highest sensitivity. Joint detection index can increase the sensitivity of the diagnosis of bone metastases, but at the same time, specificity is reduced. Therefore, two joint inspection shall be tested on a single index on the basis of improve the specificity and sensitivity.Joint inspection of these factors is more significance for bone metastases in breast cancer.The detection of cellular immune function in patients with bone metastases suggest that the decreased levels of CD4+ cells with elevated levels of CD8+, CD19+ cells may be a sign of breast cancer recurrence or deterioration.
