Object: To estimate the value of CA125 and CA15-3 in diagnosis and monitoring of breast cancer, as well as their correlativity with clinical pathological factors in breast cancer.Method: Radioimmunoassay mathod was used to determine the serum levels of CA125 and CA15-3 in 80 breast carcinoma (preoperative patients), 60 benign breast disease and 80 normal adults. The serum levels of CA125 and CA15-3 were monitored after operation, irradiation and chemotherapy.Result: 1. The serum levels of CA125 and CA15-3 in stage III-IV breast cancer were higher than benign breast disease and normal adults(P<0.05); 2. Serum CA125 and CA15-3 in stage III-IV breast cancer were higher than stage I -II (P<0.05); 3. Using cut-off levels of 54.6U/ml (CA125) and 38.6U/ml (CA15-3) we observed a sensitivity of 10% veins 16.25% and a specificity of 100% verus 100%. The sensitivity of Combination of both CA125 and CA15-3 was 20%, the specificity 100%; 4. Serum CA125 and CA15-3 were related with the size of tumor, clinical stage, axillary lymph nodes metastasis and the expression of ER, PR and c-erB-2 (P<0.05). There wasn't correlation among CA125, CA15-3 and ages, menses status, pathological type(P>0.05); 5. The levels of CA125 and CA15-3 in breast cancer after operation were lower than before operation (P<0.05); 6. Serum CA125 and CA15-3 in recovery breast cancer patients after irradiation and chemotherapy were lower than before treatments(?<0.05).Conclusion: Serum CA125 and CA15-3 is useful to diagnosis stage III-IY breast cancer , and they can be taken as the biological markers predicting tumor progression, evaluating therapeutic response and indicating prognosis in breast cancer.
