Purpose:In this experiment, the expression of PKA,CREB and PKA in the femur, kidney and hypothalamus of rats with osteoporosis were observed to explore the mechanism of osteoporosis.According to the principle of "kidney main bone" in traditional Chinese medicine,using the kidney tonifying recipe with Yougui Wan as the presentative of the intervention on osteoporosis in rats,and observe the expression of PKA,CREB and PKA in the femur, kidney and hypothalamus of rats, so we can understand the prevention and cure mechanism of the kidney in the treatment of osteoporosis.Material and Method:In this experiment, we used the method of creating the model of the bilateral ovary to reproduce the osteoporosis in rats,60 SD rats were divided into normal group, sham operation group, model group, Yougui Wan group, Fushanmei group, 14 rats in each group.Yougui Wan is experimental medicine, Fushanmei is control group,we elected the the femur, kidney and hypothalamus after 12 weeks.Bone mineral density of the left femur of the rats was measured by dual energy X-ray,so we can test the establishment of animal model of osteoporosis and drug intervention in the treatment group.ELISA method was used to test the protein content of PKA,CREB and PKA in kidney and hypothalamus of rats,And use the SPSS software to statistics and comparison to analyze the mechanism of the occurrence of osteoporosis and the mechanism of the effect of the kidney and kidney on the prevention and treatment of osteoporosis.Results:1. Bone mineral density and serum of ALP?TRAP results in osteoporosis model group:Compared with normal group, three index expression decreased,P<0.01;Compared with model group,the bone mineral density and the expression of ALP and TRAP protein in the model group were improved by each medicine group,P<0.01.2. PKA, CREB and ATF4 of bone in osteoporosis model group rats results: compared with the normal, lower PKA and CREB expression, enhanced expression of ATF4, P<0.01;compared with the model group, the treatment group can be up-regulated PKA and CREB expression and decrease ATF4 expression,P<0.01.3. PKA, CREB and ATF4 of kidney in osteoporosis model group rats results: compared with controls, the expression of PKA, CREB and ATF4 were decreased and P<0.01; compared with the model group, each treatment group could upregulate the expression of P<0.01.4. PKA, CREB and ATF4 of hypothalamus in osteoporosis model group rats results:compared with the normal, PKA, CREB and ATF4 expression were enhanced,P<0.01;compared with the model group, each drug group could reduce its expression, P<0.01.Conclusion:1. By harvesting the bilateral ovaries of rats can be successfully established the model of osteoporosis and kidney tonifying herbs can significantly increase the bone density of rats with osteoporosis.2. In normal group, the expression of PKA, CREB and ATF4 protein in the femur, kidney and hypothalamus showed that there were PKA, CREB and ATF4 signal transduction pathways in the femur, kidney and hypothalamus of the rats in the normal group.3. Decreased expression of PKA and CREB in osteoporosis model of rat femur, enhanced expression of ATF4; renal PKA, CREB decreased expression of ATF4 expression decreased, increasing expression of PKA and CREB in hypothalamus and enhanced expression of ATF4. These may is one of the pathogenesis of osteoporosis.4. Yougui Wan can improve kidney and femur of rats in the PKA, CREB, kidney ATF4 expression level, reduce the expression levels in the hypothalamus of PKA, CREB, femur and hypothalamus ATF4 and tips of tonifying kidney essence may by promoting kidney and femur of rats in the PKA, CREB, kidney ATF4 expression and inhibiting the expression of hypothalamus in PKA, CREB, femur and hypothalamus ATF4, play a role in the prevention and treatment of osteoporosis.
