Purpose: In order to explore the etiological factors and pathology of the osteoporosis,We study the expression of GPR48, CREB, BMP9 proteins in the femur, kidney,hypothalamus of the rats of osteoporosis due to kidney deficiency by using ovariectomy.To explore the adjustable function of Zuogui Decoction and its regulating mechanism.Material and Method: 70 female 3-month old SPF taken ovariectomy to establish OP rat model. The rats were divided in to 5 groups: control group, sham group, model control group, Zuogui group and Fosamax group. After 12 weeks intragastric administration with Zuogui decoction and Fosamax respectively,serum, kidney hypothalamus and femur were harvested. bone density of femur(left hind) by X-ray examination and ALP and TRACP expression in serum were for evaluating OP model establishment and the efficiency of all the above medicines. To analyze the protein expression of GPR48, CREB, BMP9 in kidney, hypothalamus and bone. Analyzed the data statistically by SPSS software to compare the difference in between the groups to demonstrate OP development and the mechanism of Zuogui decoction in preventing and treating OP.Results:1 BMD result of each group:Comparing with control group,significantly decreasing in model group(P<0.01).Comparing with model group, Zuogui group and Fosamax group are significantly increased(P<0.01), and Zuogui group are closer to control group and sham group.2 Serum ALP result of each group:Comparing with control group, significantly decreasing in model group(P<0.01).Comparing with model group,Zuogui group and Fosamax group are significantly increased(P<0.01).3 Serum TRACP result of each group:Comparing with control group, significantly increasing in model group(P<0.01).Comparing with model group,Zuogui group and Fosamax group are significantly decreased(P<0.01).4 Result of the experimental group rats' GPR48, CREB, BMP9 protein expression in bone, kinedy and hypothalamus: GPR48, CREB, BMP9 protein expression in bone.Comparing with control group,significantly decreasing in model group(P<0.01);Comparing with model group. Zuogui group and Fosamax group are significantly increased(P<0.01). GPR48 protein expression in kidney. Comparing with control group,significantly decreasing in model group(P<0.01); Comparing with model group, no difference in Fosamax group(P>0.05), Zuogui group are increased(P<0.05). CREB,BMP9 protein expression in kidney Comparing with control group, significantly decreasing in model group(P<0.01); Comparing with model group. Zuogui group and Fosamax group are significantly increased(P<0.01). GPR48, CREB, BMP9 protein expression in hypothalamus comparing with control group, significantly increasing in model group(P<0.01); Comparing with model group, Zuogui group and Fosamax group are significantly decreased(P<0.01).Conclusion:1 Using the method of ovariectomy, the rat model of osteoporosis can be established.2 The decreased level of rat with osteoporosis model GPR48, CREB, BMP9 protein expression in bone and kidney tissue and the improved level of GPR48, CREB, BMP9 protein expression in hypothalamus tissue is one of the molecular biology mechanism.3 Improving the GPR48, CREB, BMP9 protein expression in bone and kidney tissue,and reducing the GPR48, CREB, BMP9 protein expression in hypothalamus tissue is may the Zuogui decoction's mode of action with improveing the BMD of rats with osteoporosis model in cureing osteoporosis.
