Abstract/Summary:
Purpose: The pharmacodynamic study of vitexin-2”-O-rhamnoside on "acute myocardial ischemia in rats", to clarify the characteristics of effect.Material and method: The rats were randomly divided into the blank control group,Model group,fufang danshen diwan group(85mg·kg-1), low dose group(20mg·kg-1), Middle dose group(40mg·kg-1), High dose group(80mg·kg-1). Above each lavage once a day for 15 days, 1 h after the last 3 days to intraperitoneal injection of(IP) isoproterenol hydrochlorideusp(Iso)(5mg·kg-1) induced experimental acute myocardial ischemia in rats, the last injection Iso after 6 h, with 10% chloral hydrate anesthesia, draw blood from abdominal aorta. To assess vitexin-2”-Orhamnoside protective effect on myocardial injury by determination of serum myocardial enzyme [creatine kinase-MB(CK-MB), hydroxybutyric dehydrogenase(HBDH), lactate dehydrogenase(LDH)] activity. By measuring the serum lipid [high density lipoprotein cholesterol(HDL- C), low density lipoprotein cholesterol(LDL-C), triglycerides(TG)] content to assess vitexin-2”-O-rhamnoside depressing blood-fat effect.Results:1.Compared with the blank group, model group CK-MB, LDH and HBDH levels were significantly higher(p< 0.05); Compared with model group, vitexin-2”-O-rhamnoside group CK-MB, LDH and HBDH levels were significantly or extremely significantly reduced(p< 0.05); compared with fufang danshen diwan group, vitexin-2”-Orhamnoside group CK-MB, LDH and HBDH content difference is not significant.2.Compared with the control group, HDL-C model group, LDL-C content had no obvious difference, the content of TG in model group was increased, compared with statistical significance(p< 0.05). Compared with the model group, HDL-C in the high dose group increased, the difference is statistically significant(p< 0.05) and the rest of the treatment group no significant change; LDL-C content in addition to the low dose group had no significant difference, the rest of the group and the model group are significantly reduced, with statistical software SPSS have significant differences(p< 0.05); the content of TG in each treatment group was significantly lower than that of model group, and there was significant difference(p< 0.05).Conclusion: vitexin-2”-O-rhamnoside can significantly improve the serum myocardial enzyme damage in rats with myocardial ischemia in patients with coronary heart disease and reduce the CK-MB, LDH, HBDH content, lipid-lowering effects, on isoproterenol induced rat myocardial ischemia of coronary heart disease damage has significant protective effect, provides a new drug of choice for the treatment of myocardial ischemia.
