Microfluidic chip technology is a new bio-chip technology and developed in the 1990s,it's a interdisciplinary science with analytical chemistry,MEMS,computer,electronic information,materials,biology and medicine,etc.Microfluidic chip technology gave the basic operation possibility to sample preparation,biological and chemical reaction,separation and detection,which is integrated into a few square centimeters chip and complete different biological or chenical reaction,and analysis the product.In a word like move the lab to a chip.Compared with the traditional experimental method,this technology have the advantages of rapid analysis,high throughput and low sample consumption,structure miniaturization,automation,etc.With the rapid development of the technology and the continuous expansion of application areas,microfluidic technology is widely used in circulating tumor cell capture and release,cell biology research,DNA and other biosensors,cell detection and sorting,micro-droplet preparation and micro-control and other fields.In this paper,a simple Y-shape microchannel chip is used to fabricate janus droplets and manipulate the droplets to achieve monodisperse mono-layer arrangement,double-layer arrangement and multi-layer arrangement.The diffusion mechanism inside the janus droplets is studied to reduce two-phase mixing.We hope that,through this experiments,we can be controlled,high-throughput synthesis of other required janus particles.Specific work content has the following points:1.Based on the preliminary work of our laboratory and the needs of this experiment,we designed a microfluidic chip with Y-shape flow-focusing2.microchannel structure.3.Using this Y-shape structure to generation janus droplets.Respectively,by adjusting the continuous phase and dispersed phase fluid flow rate,the frequency of droplet generation and the size of droplets chould be effective controllable.4.Optimize the experimental parameters to achieve the janus droplets of mono-layer arrangement,double-layer and multi-layer arrangement.By detecting the fluorescence intensity of a single janus droplet,the internal diffusion mechanism is explored to reduce the two-phase mixing.
