Effects Of Crude Polysaccharide From Arenaria Kansuensis On Tumor Growth In Sarcoma 180 Tumor-bearing Mice

ObjectiveTo investigate the effect of Arenaria kansuensis crude polysaccharide?AKCP?on the tumor growth of S180 tumor-bearing mice.To explore the effect of AKCP on the expression level of gene and protein of proliferation and apoptosis-related factors in S180 tumor-bearing mice,and to study its possible anti-tumor effect.The mechanism provides the research basis and theoretical basis for the further separation and screening of the anti-tumor effective components of Arenaria kansuensis.MethodsS180 tumor-bearing mice model was established.Forty Balb/c mice were randomly divided into 5 groups with 8 in each group:control group,cyclophosphamide?CTX?group,and AKCP high-dose?AKCP-H?,Medium-dose?AKCP-M?,Low-dose?AKCP-L?groups.Among them,mice in the Control group were given intragastric administration of distilled water,mice in the CTX group were given intraperitoneal injection of cyclophosphamide?CTX?at 20 mg·kg^-1,mice in the AKCP-H,AKCP-M,and AKCP-L groups were given gavage at 200 mg·kg^-1,100mg·kg^-1,50mg·kg^-11 respectively.Mice were sacrificed after ten days continuous administration,then the tumor tissue blocks were completely removed under sterile conditions.Tumor was weighed and the tumor inhibition rate was calculated;The effect of AKCP on the morphological changes of the tumor tissues in each group was observed by HE staining;Expression of proliferation-related factors CyclinD1,PCNA,Ki67 and apoptosis-related factors Bcl-2,Bax of the cells in the tumor tissues were detected by fluorescence quantitative PCR;Western blot was used to detect the expressionlevelofproliferation-relatedfactorsCyclinD1,PCNAand apoptosis-related factors Bcl-2,Bax in tumor tissue.ResultThe tumor weight in CTX,AKCP-H,AKCP-M and AKCP-L groups were less than control group?P<0.05?,the tumor inhibition rate were 58.033%,25.574%,31.803%and 23.279%in these groups.HE staining showed that necrosis area and interstitial increase were occurred in tumor tissues of CTX and AKCP groups.There were no difference on mRNA expression levels of CyclinD1,PCNA,Ki67,Bcl-2,Bax,Bcl-2/Bax between control group and other groups by fluorescence quantitative PCR?P>0.05?;There were no difference on protein expression levels of CyclinD1,PCNA,Bcl-2,Bax,Bcl-2/Bax between control group and other groups by Western blot?P>0.05?.Conclusion AKCP inhibited the tumor growth of S180 tumor-bearing mice.AKCP had no effect on expression of cell proliferation?CyclinD1?PCNA?Ki67?and apoptosis-related factors?Bcl-2?Bax?in tumor tissue of S180 tumor-bearing mice.
