Objective:This project attempted to observe the "Xiusanzhen" intervention in nasal olfactory bulb ultrastructure and the midbrain nigra GFAP expression of Parkinson's disease mice induced by lipopolysaccharide(LPS).Tried to elucidate the mechanism and possibility of prevention and control of Parkinson's disease from the perspective of intervention olfactory pathway,looking forward to provide ideas and evidence for diagnosis and treatment of Parkinson's disease.Methods:60 KM male mice were randomly divided into 4 groups evenly,the blank control group(Blank),the Parkinson disease model group(Model),the "Xiusanzhen"intervention group(XSZ)and the western drug L-DOPA group(L-DOPA),each group contains 15 mice respectively.Model group?XSZ group and L-DOPA group were all build up to establish the Parkinson's model with the method of nasal perfusion of LPS,each side of the nostril was dripped 10?L one time a day,which lasting for 2 month.The blank control group was dripped with the normal saline.In order to avoid acute stress injury in the lungs,it was noted to keep the head of the gun light and slow enough during injection of the perfusion of LPS.After 8 weeks.unqualified mice were removed from each model group with 10 remaining,and 10 remaining in blank groups randomly.XSZ group were given the method of electroacupuncture to intervene.Acupoint selection:EX-HN3(2mm upper to the midpoint of two eyes),LI20(On the both lateral side of the nostrils,the border of hair and hair-less).All the acupoints were abliquely punctured 2mm with dilatational wave with 1mA's strength.Each LI20 and EX-HN3 was connected with the positive and negative pole respectively for 10 minutes'stimulation,20 minutes in total:1 time/day,5 days for a period of treatment followed with a rest for 2 days.The evaluation of curative effect was checked after 4 periods of treatment.L-dopa injection peritoneal injected once a day at exact time,5 days for a period of treatment followed with a rest for 2 days.Mice were executed after 4 periods of treatment.After that,transmission electron microscopy was used to observe the ultrastructural changes of the olfactory bulb and the changes in the expression of GFAP.The result:Ultrastructure of olfactory bulb showed that the pathological changes of the organelle and ultrastructure of the olfactory bulb in the PD group were obviously changed under the electron microscope.Compared with the blank group,The expression of GFAP had increased in PD model group(P<0.001).At the same time,it was showed that "Xiusanzhen" intervention group had not only inhibited the change of olfactory bulb ultrastructure but also reduced the expression of GFAP.(P<0.01)Conclusion:"Xiusanzhen" intervention can improve the behavioral disorders of model mice with Parkinson's disease that induced by LPS,the mechanism may be related to the function that "Xiusanzhen" could protect the olfactory bulb and reduce the expression of GFAP through the olfactory pathway.
