Objective:To research the differentially expressed plasma exosome proteins of schizophrenia comparing to the healthy people.We screened and analyzed the differentially expressed protein as potential biomarkers,which could as a simple method to diagnosis with schizophrenia or treat efficacy monitoring of drugs with schizophrenia.Method:We used quantitative proteomics TMT(tandem mas tag)connecting with Mass spectrometry technique to detect the plasma exosome proteins of 15 cases of first episode schizophrenia,42 cases of schizophrenic patents treated by medicine and 45 cases of healthy people.Meanwhile,we also had a quantitative analysis of protein and choosed the differentially expressed plasma exosome proteins of schizophrenias.Maxquant software,Uniport databae and String website were the application of bioinformatics analysis.We reviewed many literatures to screen biological markers related to schizophrenias.The proteins are validated by Parallel reaction monitoring.Results:One hundred and sixteen differentially expressed plasma exosome proteins were identified.The first episode schizophrenic patients compared with the healthy people,92 proteins were significantly up-regulated,and 6 proteins were significantly down-regulated(P<0.01).The first episode schizophrenic patients compared with schizophrenic patients with drug treatment,4 proteins were significantly up-regulated,and 22 proteins were significantly down-regulated(P < 0.01).The schizophrenic patients with drug treatment compared with the healthy people,11 proteins were significantly up-regulated,and 15 proteins were significantly down-regulated(P < 0.01).The Gene annotation analysed found differentially expressed plasma exosome proteins are mainly participating in 15 kinds f biological process,from 16 kinds of cellular component,having 6 kinds of molecular function.HP,SAP,ANP,CAM,PSMB7 were preliminary screened as candidate protein markers related to schizophrenia.There is a statistical significance between the three groups of data,p<0.01.The quantitative data of HP and SAP were basically the same as those of TMT(p<0.001)by PRM testing,while ANP was opposite(p>0.05).Conclusion:The differentially expressed plasma exosome proteins in the first episode schizophrenic patients were different from those in the schizophrenic patients with drug treatment and the healthy people.These differences may become specific plasma exosome proteins markers which could diagnose with schizophrenia easily or treat efficacy monitoring of drugs in schizophrenic patients.
