Objective:To observe the effect of methyl eugenol(ME)on the expression of aquaporin(AQP)5 in nasal mucosa of rats with allergic rhinitis and explore its significance.Methods : Wistar rats were randomly divided into the normal group,AR group,budesonide group and ME different dose intervention group(80 mg/kg group,40 mg/kg group,20 mg/kg group and 10 mg/kg group).Rats were interfered by ovalbumin(OVA)to establish the model of AR.After 1,2 and 4 weeks of administration,the distribution of AQP 5 in nasal mucosa was observed by immunohistochemistry;the expression of AQP 5 in nasal mucosa of each group was identified by Western blotting;the level of AQP 5 mRNA was determined by Real-time PCR.Results:AQP 5 is mainly located in the menbrane and cytoplasm of nasal glandular epithelium and ductal epithelial cells.In comparison with the normal group,the expression of AQP 5 in nasal mucosa of AR rats was decreased at the level of protein and mRNA(P < 0.05).The expression of AQP 5 protein and mRNA in nasal mucosa of all treatment groups were higher than that of AR group.At the protein level,there was no significant difference in the expression of AQP 5 in nasal mucosa after one week of intervention with ME 20 mg/kg compared with the normal group and budesonide group.(P > 0.05),however,comparison with AR group,the difference is statistically significant(P < 0.05).At the level of mRNA,after one week intervention,the expression of AQP 5 mRNA in nasal mucosa of ME 20mg/kg group was not statistically different from that of nomal group.(P > 0.05).Conclusion:The expression of AQP 5 in nasal mucosa of AR rats was lower than that in normal rats,but the expression of AQP 5 increased after treatment with ME or budesonide.It is speculated that ME may reduce the degree of glandular edema of the nasal mucosa and reduce glandular secretion by up-regulating the expression of AQP 5,thereby alleviating symptoms such as nasal itching,sneezing,and runny nose in allergic rhinitis.
