Objective: To observe the effection with the uric acid on the expression and function of Transforming growth factor-?1(TGF-?1)and the proliferation ability of osteoblast-like cells(MG-63)in vitro.Methods: The osteoblast-like cells(MG-63)in good condition were divided into four groups: the control group by adding complete medium and the three treatment groups by adding different concentrations of uric acid(0.2mmol/L,0.4mmol/Land 0.8mmol/L)in osteoblast inducing media.Cell morphology was ascertained under an inverted microscope after fourteen days of induction.After seven days of induction,cell proliferation ability was detected by alkaline phosphatase activity;Cell proliferation was detected by CCK-8 method,which was induced by cell culture to 3d,5d,7d and14 d,respectively.The expression of transforming growth factor-?1(TGF – ?1)m RNA was identified by reverse transcription PCR(RT-PCR).Results:(1)Cytology under inverted microscope showed that cell number incresed obviously with the increse of uric acid concention.(2)Alkaline phosphatase activity were prompted by cell in culture medium with osteogenesis culture medium and the uric acid intervention culture medium: cell proliferation strengthened as uric acid concentration increased.The comparison between each experimental group and control group or the comparison between each experimental group has statistic significance(p< 0.05).(3)Cell counting kit-8(CCK-8)method showed that OD value of the uric acid groups is significantly higher than that of the control group by the comparison between each experimental group and control group or the comparison between each experimental group.We found that OD value is higher by the increase of uric acid concentration.The difference is statistically significant.(P<0.05).(4)RT-PCR results:the expression of TGF-beta1 m RNA was increased with the increase of uric acid concentration after seven days and fourteen days.Compared between each group,the difference significant was statistically.(P<0.05).Conclusions:(1)Uric acid can promote the osteoblast-like cells proliferation in vitro confirmed by cell morphology,ALP activity assay and CCK-8 assay,with a characteristic of concentration-dependent and time-dependent.(2)Uric acid can promote the osteoblast-like cells proliferation in vitro,by the method of up-regulating the expression of TGF-?1 m RNA,which is dependent on concentration and time.
