In the last century,there were few reports about lung cancer.However,in all malignant tumors,it has becomeing the Number one male morbidity and mortality,and the second highest among women.Lung squamous cell carcinoma is a common pathological type of lung cancer.The early symptoms are often not obvious and are often overlooked,so most patients have entered the middle and late stages when they are diagnosed.During the process of cell mitosis,if the synthesis of tubulin is blocked,it will eventually lead to death of cells.Tumor treatment fields(TTFields)is based on a specific low-intensity,intermedium frequency electric field(100-300 kHz),it can prevent the formation of spindle microtubules in the mitosis of tumor cells and the separation of intracellular organelles during cell division,and can also induce apoptosis in the dividing phase to achieve the purpose of anti-tumor.Based on the above background,this experiment intends to observe the effect of TTFields on ?-tubulin synthesis in lung squamous cell carcinoma cells(SK-MES-1),the ability of inducing apoptosis and the effect on their invasive ability through in vitro experiments.A total of 4 groups of experiments were set up,ie no electric field group,100 kHz,150 kHz and 200 kHz.The cells were cultured for 6 hours and microscopically confirmed that the cells grew normally.After that,the above electric field conditions were applied and the cells were continuously electrified for 72 hours.Then the cells were collected and subjected to Western Blotting to detect the expression of ?-tubulin.Flow cytometer was used to analyze cell apoptosis and Transwell assay was used to detect the invasion ability of tumor cells.The results of the four groups of experiments were as follows: The expression of?-tubulin in the 150 kHz group was the lowest,and the difference was statistically significant(P<0.05).The other three groups had no significant differences among them(P>0.05).The 150 kHz group had the most significant effect on inducing apoptosis and tumor cell invasiveness,which was statistically significant compared with the other three groups(P<0.05);the 100 kHz and 200 kHz groups were relatively poor,and there was no significant difference between the two groups(P>0.05).They were still statistically significant in the non-electric field group(P<0.05).TTFields act on lung squamous cell carcinima,interferes with the assembly ofspindle microtubules by inhibiting the synthesis of ?-tubulin,then preventing mitotic progression and leading to cell death.At the same time,TTFields can also reduce the invasiveness of tumor cells,destroy the structure of tumor cells,and induce tumor cell apoptosis.
