BackgroundOral squamous cell carcinoma is one of the most common malignant tumors in clinic.The incidence of OSCC is increasing year by year,most of OSCC is treated with surgery-based comprehensive treatment,the radical operation of OSCC has greatly damaged the patient's face,which seriously affected the patient's quality of life.Therefore,the chemical prevention and treatment of OSCC has become a new research hotspot.Prostaglandin E2(PGE2)is a proinflammatory mediator and belongs to the family of bioactive lipids.A large number of studies have found that the expression of PGE2 is upregulated in different tumors,and PGE2 can promote the occurrence and development of tumors,including promoting tumor cell proliferation,tolerance of apoptosis signals,increased angiogenesis,enhancement of invasion and metastasis ability,and evasion of host immune response.It is mainly produced by cyclooxygenases(COX-1 and COX-2)and microsomal prostaglandin E synthase 1(mPGES-1).COX-2 has been targeted for research in many cancers,including oral squamous cell carcinoma,breast cancer,and prostate cancer.In recent years,a large number of studies have confirmed that long-term use of non-steroidal anti-inflammatory drugs such as COX-2 inhibitors has serious side effects on the gastrointestinal tract and cardiovascular system.Therefore,we are motivated to explore downstream mPGES-1 and its related mechanisms of action.Therefore,in order to further explore the role of mPGES-1 in the occurrence and development of oral cancer,this study used different concentration gradient mPGES-1 inhibitor CAY10526 to act on tongue cancer cell line CAL27,and explored its effect on biological characteristics.Aim1.Clarify the expression of mPGES-1 in oral cancer.2.To explore the effect of mPGES-1 inhibitor CAY10526 on the proliferation,migration and invasion of tongue cancer cells.3.To explore the effect of mPGES-1 inhibitor CAY10526 on the apoptosis and cell cycle of tongue cancer cells.Method1.Use the TCGA database to detect the expression of mPGES-1 in normal tissues and tumors.Immunohistochemical staining was used to detect the expression of mPGES-1 in oral cancer and adjacent tissues.qRT-PCR and Western Blot were used to detect the expression of mPGES-1 in CAL27 and SCC-15.2.Use CCK-8 to detect the effect of mPGES inhibitor CAY1056 on the proliferation of tongue cancer cells,and use the scratch test and Transwell migration and invasion test to detect the effect of CAY1056 on the migration and invasion ability of tongue cancer cells.3.Flow cytometry was used to detect the effect of CAY1056 on the apoptosis and cell cycle of tongue cancer cells,and Western Blot was used to detect the expression of apoptosis and cycle-related proteins.Result1.TCGA database analysis showed that mPGES-1 was differentially expressed in head and neck squamous cell carcinoma,and its expression increased.The expression of mPGES-1 in oral squamous cell carcinoma is higher than that in adjacent tissues,and the expression of mPGES-1 mRNA in tongue cancer cell line CAL27 is higher than that of SCC-15.2.CAY10526 can inhibit the proliferation of tongue cancer cells,and the proliferation ability of cells in 50?M group CAY10526 is significantly reduced;CAY10526 can inhibit the migration of tongue cancer cells,and the effect of CAY10526 in 50?M group is the most significant,the number of migrated cells is 37.00 ± 7.81 Significantly lower than the control group 150.3 ± 2.96;CAY10526 can inhibit tongue cancer cell invasion,and the 50?M group CAY10526 has the most significant effect,the number of invasive cells is 30.33 ± 7.31,significantly lower than the control group 110.00 ± 5.13.3.CAY10526 can promote the apoptosis of tongue cancer cells,and the effect of CAY10526 is the most significant in the 50?M group,the apoptosis rate is(16.90± 0.45)%,lower than the control group(4.34 ± 0.29)%;In the G1 phase,the CAY10526 blockade effect was the most significant in the 50?M group.The percentage of cells in the G1 phase was(92.11±0.83)%,which was significantly higher than the control group(44.24±0.36)%.Conclusion1.mPGES-1 is highly expressed in tongue cancer cells.2.The mPGES-1 inhibitor CAY 10526 can inhibit the proliferation,migration and invasion of tongue cancer cells in a concentration-dependent manner.3.The mPGES-1 inhibitor CAY 10526 promotes apoptosis of tongue cancer cells by down-regulating the expression of BCL-2 and up-regulating the expression of BAX,and blocks the cells in G1 phase by down-regulating the expression of Cyclin D1 and CDK4.
