Background: AIM2 gene is located in the cytoplasm,can feel the exogenous double-stranded DNA(ds DNA),and initiate a series of inflammatory immune responses.Many studies have found that AIM2 is not only associated with inflammatory responses,but also with tumor occurrence and development.Recently studies have found that AIM2 gene can inhibit colorectal tumors in mice,but in the process of the specific mechanism and the clinical value of AIM2 gene is not yet clear,the domestic and foreign relevant AIM2 genotype and colorectal cancer clinical research.Objective: This study by comparing the colorectal cancer patients with cancer three different tissue-cancer tissue,adenoma tissue and normal tissue,as well as a lymph node cancer cells infiltration(N+)in patients with positive lymph nodes,and without lymph node cancer cells infiltrating lymph nodes(N-)patients AIM2 gene expression analysis AIM2 genes expressed in different tissues in cancer patients and its relationship with clinical pathology data.Methods: 1.In November 2016 to sep 2017,we collecting gastrointestinal surgery surgical specimens of colorectal tumor surgery,which needs to have cancer tissue at the same time,in each specimen gland tumor tissue and normal tissue,and with complete clinical pathological information.2.Getting cancer organizations,more than 10 cm away from the tumor patients with normal tissue,adenoma groups and the N+ patients cancer cells infiltrating lymph nodes,lymph nodes,N-patients on AIM2 gene specificity immunohistochemical staining,the immunohistochemical room physician help downlink result determination,classification comparison and analysis the organization AIM2 dyeing results,and the relationship between the clinical pathological data.3.Data analysis was performed using SPSS17.0 statistical software for the clinical pathological data and AIM2 staining score of the collected patients.4.Involving the immunohistochemical method: The research adopts the method of polymer(Labelled dextran polymer,LDP),is using horseradish peroxidase(horseradish peroxidase,HPR)tag on the glucan polymer,and with the second antibody preparation into combination,formed En Vison two antibody,when the first antibody and antigen specific substrates were detected after combination,joined En Vison two resistance,forming enzyme-polymer compound-the second antibody complex.The specific color reaction was detected by Diaminobenzidine(DAB).Results:The study included 71 patients,including 53 males,about 74.6%;including 18 females males,about 25.4%.The mean age of all the patients was 63.84 ± 13.21 years old,22 patients with lymph node metastasis,about 31.0%,and 49 patients without lymph node metastasis,about 69.0%.1.AIM2 specific staining showed that main AIM2 gene expression in colorectal mucosa epithelial cells in the cytoplasm,specific immune group dyeing with light yellow,brown and tan,which in normal tissue adjacent to carcinoma and adenoma tissues,relatively concentrated in mucosal epithelium or opposite sex glands hyperplasia gland in the cytoplasm,secondly in lymphoid tissue,expressed less other stromal cells.In cancer tissues,the expression is diverse.2.In 71 cases of surgically resected specimen,cancerous tissue AIM2 didn't express or lower expression(-or +)34 cases(47.9%),adenoma tissues AIM2 didn't express or low expression in 10 cases(14.1%),AIM2 no expression in normal mucosa tissues or low expression in 2 cases(2.8%).According to SPSS analysis results,the expression of AIM2 gene was suggested to be the highest in normal tissues,and the lowest in adenoma tissues(P<0.05).3.In 18 patients with lymph node metastasis(N+),each patient had an average of 2.72 ± 1.27 positive lymph nodes.There were 49 patients without lymph node metastasis(N-),and 12.22± 5.79 lymph nodes were detected in each case.In the positive lymph nodes of N+ patients,the low expression rate of AIM2 was higher than that of N-patients(55.6% vs 22.4%,?2 = 6.705,P=0.010).The low expression rate of AIM2 in patients with N+ patients was higher than that in n-patients(66.7% vs 34.7%,?2 = 5.482,P = 0.019).4.According to the expression of AIM2 in colorectal cancer and adenoma tissue,the patients were classified according to grade respectively.In colorectal tissue,AIM2 with TNM staging of the tumor patients(Z =-2.826,P = 0.005),tumor infiltration depth(Z =-2.915,P = 0.004),lymph node cancer cells infiltration or not(Z =-2.435,P = 0.015),the CEA preoperatively(Z =-2.093,P = 0.036)and clinical pathological grouping index differences statistically significant.In the patients with gender(Z =-0.342,P = 0.732),age(Z =-1.226,P = 0.220)and tumor size(Z =-1.739,P = 0.082),tumor site(?2 = 5.345,P = 0.069),tumor pathological type(?2 = 5.905,P = 0.052),pathological differentiation(?2 = 4.592,P = 0.101)group,there was no statistically significant difference.5.In adenoma tissues,AIM2 in adenoma atypia group differences statistically significant(Z =-2.539,P = 0.011 < 0.05),and the expression level of AIM2 was decreased as the heterogeneity increased.In the patients with gender(Z =-0.055,P = 0.956),age(Z =-0.844,P = 0.399),and the size of the adenoma(?2 = 1.106,P = 0.575)the location of adenoma(?2 = 2.108,P = 0.550),the pathological type of adenoma(?2 = 1.315,P = 0.518),no statistically significant difference on grouping(P > 0.05).Conclusion: 1.AIM2 mainly expressed in the cytoplasm of epithelial cells of colorectal,among them the highest expression in normal tissue,adenoma tissue expression is reduced,the lowest in the cancer tissue,suggesting AIM2 gene may be colorectal cancer suppressor genes,and through normal tissue-cancer-adenoma tissue the development route.2.In patients with N+,the expression of AIM2 in cancer tissues and positive lymph nodes was lower than that of N-patients,indicating that patients with low expression of AIM2 gene were more likely to have lymph node metastasis.3.Patients with low expression of AIM2 in colorectal cancer tissue,the tumor clinical pathological stage was late and the prognosis was worse.It is suggested that AIM2 may be a prognostic indicator for patients with colorectal cancer.
