Objective: Sporotrichosis is a chronic infectious disease caused by the infection of skin,subcutaneous tissue,mucous membrane and local lymphatic system by Sporothrix schenckii complex,having varied clinical manifestations,and the main skin lesions are chronic inflammatory granulomatous lesions.Studies have shown that the host's recognition and response to the pathogenic microorganism is highly dependent on the immune system.Dendritic cell is currently known as the most powerful professional antigen presenting cell and is regarded as the bridge of innate immunity and adaptive immunity.In this study,the dynamic expression of TNF-?,IFN-?,IL-4 and other inflammatory factors in the infection model of Sporothrix schenckii complex was detected to provide the basis for the mechanism of the inflammatory reaction in Sporotrichosis.Methods: In order to study if the host secretes TNF-?,IFN-? and IL-4 after infecting Sporothrix schenckii complex,we use the strains isolated from the patient's tissue to stimulate the dendritic cells in mice.Then we detect the dynamic expression of TNF-?,IFN-? and IL-4 in the supernatant by ELISA and Western blotting.Result: In the model of Bone marrow-derived dendritic cells stimulated by Sporothrix schenckii complex,TNF-? can be produced at early stage,but inflammatory factors such as IFN-? and IL-4 are hardly secreted.Data showed at the same stimulation time when the bacterial concentration increased,the secretion of TNF-? also increased.Under the same concentration of liquid,the secretion of TNF-? gradually decreased in 6h-24 h and increased in 24h-72 h.Conclusion: This study shows that Sporothrix schenckii complex stimulates mouse myeloid dendritic cells can secrete inflammatory factors like TNF-?,hardly secrete inflammatory factors like IFN-? and IL-4.The data show that the TNF-? produced by Bone marrow-derived dendritic cells stimulated by Sporothrix schenckii complex is time and concentration dependent,which may play an important role in the early inflammatory response of the disease,and is an important inflammatory factor in the innate immune response of Sporotrichosis.
