With the wide use of the anti-malignant tumor drug , immunosuppressive drugs and the development of broad-spectrum antibiotic , organ transplanting and other intervenal treatment , and with the spread of the AIDS , fungal infection has become the hidden killer to people's health. In recent years , fungal infection appeared many new behaviors , increasing incidence rate , death rate and to be disabled . It has become very important that early diagnosis and specifical treatment for fungal infection to save the patient and rise the relieve quality.The classification of fungi is a huge system engineering . The traditional fungi taxology depends on the morphological , physiobiochemistry and antigen construct feature. However , since there are so many species of fungi and obvious individual polymorphisms that it can not reflect the entity characteristics of fungi . And it will cause massiness workload , consumeing long and promiscuous conclusion.In the recent thrity years , with the rapid development of molecular biology , fungal taxonomy has entered the new era from phenotype to genetype . There are many moleculartechnologies , such as fungi genome DNA G+C mol% , nucleic acid hybridization , chromosome electrophoresis (EK), ribose type , genome DNA restrict fragment length polymorphism (RFLP),solobilital protein electrophoresis , multi-enzyme elesctrphoresis (MEE)and ribosomal RNA genes sequential analysis.These techonologies have differences in the resolving power and typing.Ribosomal RNA genes analysis is believed to be one of the best indicators that could reflect the genetic affinity among species. In the past , the emphasis in the ribobomal RNA genes focused on the conservative sequence . But now , there are more and more investigater founding that the spacer regions , as ITS and IGS . The main means are probing hybridization , PCR, RFLP and sequence analysis . And sequence analysis is believed to be one of the most directness and clear targets.Sporothrix schenckii is the causative agent of sporotrichosis , which affects primarily the skin , subcutaneous tissue , lymph vessels , lymphnodes and rarely , visceral organs . Although fixed lesions are common , disseminated infection by sporothrix schenckii can be found occasionally . The clinical manifestations of sporotrichosis are related to immune status of theindividual . The conventional taxonomy of fungi has been mainly baseing on morphology . The problem of sporothrix schenckii morphotyping is that S.schenckii can not be sub-divided within the species . In this study , we investigated the mycologic feathres and variations in gene sequence of a sporothrix strain which caused disseminated cutaneous sporothrichosis ,with exploreing a simple and rapid method for identification of Sporothrix schenckii.Materials and Methods: Three clinical isolated of S.schenckii from patients with different clinical manifestations were included in this study . The sporothrix strain which caused disseminated cutaneous sporotrichosis was identified by routine mycologic and polymerase chain reaction(PCR) techniques . Ribosomal conserved sequence.Results: The clinical isolated causing disseminated cutaneous sporothrichosis was identified as S.schenckii by routine morphological and PCR techniques.Conclusion: Sporothrix schenckii can be easily differentiated from other pathogen by PCR .The ITS sequences were highly conserved among S.schenckii strains , which suggests that this region might be suitable as the target gene for designing species-specific primers orprobes .
