Objective 1.To establish a stable animal model of posterior limb and knee joint contracture in Wistar rats,so as to establish an animal model platform for the study of joint contracture.2.To measure the range of joint motion after intervention of model animals into groups,to explore the effect of KERUTI on joint motion in rats with joint contracture.Methods: 60 healthy male Wistar rats with SPF grade were randomly divided into two groups: 12 normal rats and 48 model groups.The rats' knee joint of model group were immobilized for three weeks to establish animal model of joint contracture.After the successful establishment of the model,the successful model was randomly divided into 4groups: model group,KERUTI group,exercise group and KERUTI combined exercise group,model group did not receive any treatment,the other three groups were treated with different intervention program for 4 weeks,all rats were taken X-ray under anaesthesia to measure the joint motion.The expression of TGF-? 1 in articular fluid was detected by extraction of articular fluid,and synovial tissue was taken from synovium by euthanasia for pathological section.Results:One rat in the model group suffered from anaesthesia intolerance and one rat suffered from femoral fracture in the course of model making.Two rats suffered from femoral fracture in the first week after operation.The remaining 44 model rats were all in accordance with the following observation indexes,and the success rate of model making was 91.67%.After four weeks of intervention,the comparison of joint motion in each group: compared with the model control group and KERUTI exercise group,the knee motion of the model rats increased(P<0.05),but there was no significant difference between the exercise group and the control group.Compared with the exercisegroup and the KERUTI exercise group,the knee joint motion of the experimental animals in the KERUTI group and the KERUTI exercise group also increased(P<0.05).Conclusion: 1.The rat model of knee joint contracture was successfully established,and the success rate of the model was high,and the model was stable and reliable.It can lay a good animal model for the study of joint contracture in the future.KERUTI can enlarge the range of joint motion in rats with joint contracture.
