Objective:? The main chemical constituents in parochetus communis ethanol extracts(PCE)were analysed rapidly.?The quality control method for parochetus communis were established.?The toxicity and safe oral dose of PCE were evaluated.?The effects of PCE on blood pressure in renal hypertensive rats(RHR)were investigated Methods:?The mass fragments of main compounds in PCE were determined by LC-ESI-MS method and the possible chemical structure of the constituents in PCE were analysed according to the characteristic ion fragments and compared with reported references.?The method of TLC identification,HPLC content determination and HPLC fingerprint for parochetus communis were established.?The toxicity and safe oral dose of PCE were evaluated by acute toxicity test in mice.?RHR model were established under two-kidney one-clip(2KIC)method and the blood pressure was measured using non-invasive blood pressure measuring device for rats 2 weeks after 2K1C operation.Only hypertensive rats(SBP above 150 mmHg)were used in the experiment.RHR were divided randomly into six groups:sham,model,PCE-low(100 mg/kg),PCE-medium(200 mg/kg),PCE-high(400 mg/kg)and captopril(30 mg/kg).Blood samples were collected after 8 weeks of treatment and the Ang?,ET,RE,NO and AP-12 levels,which are importang biomarkers relavant to RAS,were measured.Results:?Possible chemical structure of 9 flavonoid glycosides in PCE were identified and MS datas of 12 unknown chemical constituents were determined.?The method of TLC identification,HPLC content determination and HPLC fingerprint for parochetus communis were established using two main flavonoid glycosides as marked components,including rutin and kaempferol-3-O-rutinoside.?The result of acute toxicity test showed that the maximum concentration and maximum dose of PCE did not show any death or other unusual diseases on mice and the maximum oral dose of PCE was determined to be 40 g/kg by maximum dosage experiment.?Compared with model,systolic blood pressure(SBP)and diastolic blood pressure(DBP)in PCE-low,PCE-middle,PCE-high and captopril groups were all decreased as prolongation of treatment time and PCE showed dose-dependent effect on blood pressure in RHR;The determination results of RAS-relavant biomarkers showed that the levels of Ang?,ET,RE,AP-12 in PCE-low,PCE-middle,PCE-high and captopril groups were decreased and the level of NO was increased after 8 weeks of treatment compared with model.Furthermore,PCE has dose-dependent effect on these biomarkers.Conclusion:?The established LC-ESI-MS analysis method is fast,simple and can be used for the qualitative analysis of the main chemical components in parochetus communis.?The established quality control method is stable,reliable and reproducible,which can reflect the quality of parochetus communis and can be applied to the authenticity identification and quality evaluation of this medicinal material.?PCE showed no toxicity and safe for oral administration.?PCE can reduce the blood pressure of RHR,and its antihypertensive effects are related to RAS.
