Objective: Objective To observe the repairing effect of Periplaneta americana extract on rat skin wound,and to preliminarily explore the molecular mechanism of Periplaneta americana extract on wound healing in rats.Methods: 75 SD rats were used to establish full-thickness skin wound model of the back skin by skin acute incision method.were randomly divided into 5 groups: model group,Jingwanhong group,Periplaneta americana extract(PAE)high,medium and low treatment group.Jingwanhong group were given Jingwanhong ointment 0.1g,PAE high,middle and low group were given 250 mg/g,125 mg/g,62.5 mg/g,smear the wound,once a day,continue for 14 days,the model group were not given any treatment,respectively in the model after third days,seventh days and fourteenth days were recorded and wound healing using Image J,calculate the wound area image analysis software,to observe the morphological changes of granulation tissue with HE staining,Masson staining and growth were observed at different time points of the collagen immunofluorescence each time in each group of granulation tissue macrophages in the quantity change of VEGF,bFGF protein expression was detected by Western blot method in granulation.Results:1.the wound healing rate results show: compared with the model group,in third,seventh days after trauma,The wound healing rate of each treatment group was higher than that of the model group,The wound healing rate in the middle group of PAE was significantly increased.It has significant difference(P<0.05).in Fourteenth days,compared with the model group,the wound healing rate in the PAE middle group was obviously improved.It has significant difference(P<0.05).2.HE staining observed the growth of wound: in each time period,the treatment group was better than the model group,the model group grew slowly,the neovascularization,hair follicle,sweat gland cells and ducts in each treatment group were faster than those in the model group,among the treatmentgroup,especially the PAE middle group,the new blood vessels,hair follicles,sweat gland cells and ducts grew faster.3.Massn staining showed: compared with the model group,on the third,seventh day after injury,the expression of collagen fibers in wound tissue of each treatment group showed an increasing trend,The expression of collagen fibers in the middle group of PAE was significantly increased,it has significant difference(P<0.05).Compared with the model group,the expression of collagen fibers in wound tissue of each treatment group showed a decreasing trend,it has significant difference(P<0.05).4.immunofluorescence assay results showed that,compared with the model group,on the third,seventh day,The number of CD68 positive cells in each treatment group increased,it has significant difference(P<0.05),compared with PAE groups,PAE middle group was significantly increased,it has significant difference(P<0.05),on the fourteenth day,The number of CD68 positive cells in each treatment group showed a decreasing trend,it has significant difference(P<0.05).5.(1)Compared with the model group,on the third,seventh days,expression of VEGF showed an increasing trend in each treatment group,the expression of VEGF in the middle group of PAE was significantly increased,it has significant difference(P<0.05),on the fourteenth days,compared with the model group,the expression of VEGF in the middle group of PAE was significantly reduced,it has significant difference(P<0.05),(2)Compared with the model group,on the third,seventh days,expression of bFGF showed an increasing trend in each treatment group,the expression of bFGF in the middle group of PAE was significantly increased,it has significant difference(P<0.05),on the fourteenth days,compared with the model group,the expression of bFGF in the middle group of PAE was significantly reduced,it has significant difference(P<0.05),Conclusion: This study proves that:(1)Periplaneta americana extract is a good dosage form for repairing skin wounds.(2)the extract of Periplaneta americana has an obvious promoting effect on the repair of skin wound in rats.The mechanism may be related to regulating the expression of macrophage,VEGF and bFGF.
