| Objective: 1.To prepare SFGP blend membranes, to study the optimal dosage proportion and membrane method in line with tissue medical dressing requirements of the materials through representation observation.2. To perform biological compatibility experiment and animal trauma experiment by using SFGP blend membranes and to conduct preliminary evaluation of its feasibility and security as wound dressing.Methods: 1. Prepare SFGP blend membranes: Calcium chloride-alcohol- water system(CaCL2: C2H5OH: H2 O = 1: 2: 8) was dissolved silk fibroin. About 5% SF solution will be obtained after dialysis and concentration. Prepare G solution by dissolving gelatin into periplaneta americana extract(W11-a12)solution. Mix the two in different proportions,cross linking and plastics, standing to bubble, laying, oven dried to obtain the desired SFGK blend membranes.2. Characterization of materials: to characterize the SFGP blend membranes by SEM、Air permeability、Water absorption、Swelling ratio.3. Biocompatibility detection of the blend membrane: to experiment on SFGP blend membranes by cell culture experiments, MTT cytotoxicity test and hemolysis test.4. Perform experiment on trauma of animals by using SFGP blend membranes: to establish animal wound model, using SFGP blend membrane as wound dressing, compared to day3, day5, day7, day9,day11 and day13, to observe local wound healing, infection of the surrounding skin. Perform HE staining and Masson staining at day3, day9 and day13 to check the granulation tissue and healing of the wound.Results: 1. SFGP blend membranes stents with different proportion will be obtained after mixing of silk fibroin 、 gelatin and periplaneta americana extract(W11-a12)solution at different proportion ratio.2. To complex the characterization test results, found that blend membrane of the silk fibroin solution and gelatin mixed solution with 4:6mixed in 60 ℃ degrees and dry 8h has better air permeability, water absorption and swelling ratio.3. Cell culture experiment showed that SFGP blend membrane can promote cell growth, the MTT experiment showed that SFGP blend membrane was non-toxic, the hemolysis rate of SFGP blend membranes was 2.32%, less than 5% of the provisions of the state, in line with national standards.4. Animal trauma experiments show that the blend membranes canpromote wound healing.Conclusion: 1.Mixing silk fibroin solution、 gelatin solution and periplaneta americana extract( W11-a12) solution, were successfully prepared with stable structure and excellent characterization of blend membranes.2. SFGP blend membrane has good biocompatibility and can promote the wound healing. |
