Purpose:To study the therapeutic effect of baiying decoction on the radiation pneumonitis in rats,to explore the possible mechanism of baiying decoction for the treatment of radiation pneumonitis from the expression changes of related cytokines in lung tissue.Methods:42 male Sprague-Dawley rats were randomly divided into 5 groups: 6 rats in the blank control group(intragastric administration of normal saline 7 days before and after irradiation);9 rats in the model control group(intragastric administration of normal saline 7 days before and after irradiation);9 rats in the positive control group(intragastric administration of normal saline 7 days before and after irradiation;intraperitoneal injection of 150mg/kg amifostine 30 min before irradiation);9 rats in the low dose Chinese medicine group(intragastric administration of low dose baiying decoction 7 days before and after irradiation);9 rats in the high dose Chinese medicine group(intragastric administration of high dose baiying decoction 7 days before and after irradiation).Except for 6 rats in the blank control group,the rest rats were received the single irradiation of 6MV X-rays by means of AP-PA fields with a dose of 18 Gy.The rats were sacrificed two weeks later and lung tissues were preserved.The weight of rats during the irradiation,the body weight and the wet weight of the lungs at the time of death,HE staining of lung tissues,protein expression of TGF-?1,Smad2,P-Smad2,Smad3,P-Smad3 and Smad7 which were detected by immunoblotting were all recorded.Results:1.Body weight gain: During the two weeks after radiation,the weight of rats in each radiation group was apparently lower than those in the blank control group(P<0.01);the weight of rats in the positive control group,low dose Chinese medicine group and high dose Chinese medicine group was obviously higher than those in the model control group(P<0.01);while the rats' weight in the high dose Chinese medicine group was higher than the low dose Chinese medicine group(P<0.05)and was evidently higher than the positive control group(P<0.01);and there was no significant difference between the low dose Chinese medicine group and the positive control group(P>0.05).2.Lung coefficient: The lung coefficient of rats in each radiation group was significantly higher than those in the blank control group(P<0.01),lung coefficient of rats in the positive control group,the low dose Chinese medicine group and the high dose Chinese medicine group was obviously lower than the model control group(P<0.01);lung coefficient of rats in the high dose Chinese medicine group was lower than those in the low dose Chinese medicine group and the positive control group(P<0.05);there was no significant difference between the low dose Chinese medicine group and the positive control group(P>0.05).3.Lung tissue inflammation scores: Lung tissue inflammation score of rats in each radiation group was significantly higher than those in the blank control group(P<0.01);lung tissue inflammation score of rats in the positive control group and the low dose Chinese medicine group was lower than the model control group(P<0.05),and lung tissue inflammation score of rats in the high dose Chinese medicine group was significantly lower than the model control group(P<0.01),and there was no obviously difference among the positive control group,the low dose Chinese medicine group and the high dose Chinese medicine(P>0.05).4.Related cytokines expression in lung tissue: TGF-?1?P-Smad2?P-Smad3 expression in lung tissue of rats in each radiation group was significantly higher than those in the blank control group(P<0.01);and TGF-?1 ? P-Smad2 ? P-Smad3 expression in lung tissue of rats in the positive control group,low dose Chinese medicine group and high dose Chinese medicine group were significantly lower than the model control group(P<0.01);but there was no significant difference among the three groups(P>0.05).The expression of Smad7 in the lung tissue of the rats in the positive control group,low dose Chinese medicine group and high dose Chinese medicine group was significantly higher than those in the model control group and the blank control group(P<0.01),but there was no significant difference among the three groups(P>0.05).Conclusions:1.Baiying decoction has a therapeutic effect on radiation pneumonitis in rats.It has obviously effect on improving the weight gain,lung coefficient and inflammation score of lung tissue in rats with radiation pneumonitis.2.High dose baiying decoction,on the aspect of improving the quality of life and increasing body weight,was excel the low dose baiying decoction and amifostine.3.The therapeutic effect of baiying decoction on rats with radiation pneumonitis may be achieved by regulating TGF-? 1/Smads signal pathway,that is,inhibiting the transduction of TGF-? 1 signal in cells.
