Objective: To explore the effect of Baicalin on expression of connexin 36 in Striatum area and Brain Cortex of 6-OHDA induced Parkinson's(PD)model rats and its significance.Methods: 84 healthy Sprague-Dawley(SD)male rats were randomly divided into 6 groups included normal control group,PD model group,PD+positive control group(Madopar group),and PD+baicalin low dose,middle dose,high dose.Except the normal control group,other groups were injected with 6-OHDA in the double target of the right striatum area and the medial forebrain bundle area by microinjection with brain stereotaxic.After 3 weeks,inject apomorphine(0.5Mg/Kg)to establish PD model.On the basis of model,positive control group(Madopar 0.125g/kg/d),baicalin low dose group(50mg/kg),middle dose group(100mg/kg),high dose group(200mg/kg),normal control group and PD model group(same dose of saline)intervention.In this study,immunohistochemical and western blot used to detect connexin36 and TH in cerebral cortex and striatum,and try to investigate the expression of Cx36 in cerebral cortex and striatum and the effect of baicalin on Cx36.Results: Immunohistochemical staining and western blot showed that the number of TH positive neurons and the expression of Cx36 in PD model group were lower than those in the other 5 groups(P < 0.05),and the difference was statistically significant Compared with PD model group(P<0.05),baicalin had an upregulation effect on Cx36 expression,but there was no significant difference between each dose(P>0.05).Conclusions: The expression of Cx36 in cerebral cortex and striatum of PD model rat was decreased compared with the normal control group.Baicalin significantly up-regulated the expression of TH and Cx36.Therefore,it was initially considered that baicalin may have protective effect on dopaminergic neurons,and this feature may by the participated of Cx.
