Objective: To investigate the effects of baicalin on behavioral changes in Par-kinson's disease(PD)rats and the expression of gap junction protein 43(Cx43)i-n astrocytes(ASTs).METHODS: The rat right striatum was localized using a brain stereotaxic apparatus,and PD rat model was prepared by unilateral two-site in-jection of low dose 6-hydroxydopamine(6-OHDA).Grouping: Normal group(without special treatment),and PD rats successfully induced by modeling were rando mly divided into PD group(normal saline gavage)and Mediba group(Medopa ga-vage,125 mg/Kg),Baicalin low,medium and high dose(gastric administration,d-oses were 500mg/kg,100mg/kg,200mg/kg)a total of 6 groups,10/group,drug i nt-ervention for 28 days.After the completion of the intervention,behavioral tests were performed to observe the changes in the number of circling rats.Immunohi stochemistry was used to observe the expression of tyrosine hydroxylase(TH)cell s,and Western Blot and immunohistochemistry were used to observe the expressi on of Cx43.Results: Compared with PD group,there was no signifycant differenc e i-n behavioral testi-ng between low dose group and low dose baicalin group(P>0.05),Baicalin high dose group were significantly reduced number of turns,beh avior-ral changes signifycantly,the symptoms eased,compared with the PD group ther-e was astatistically significant difference(P<0.01);compared with the admin istrat-ion of baicalin,high dose There was no significant difference between the t wo g-roups(P>0.05).Compared with the baicalin highdose group,there was no significant change in the number of rotateons of the rats in the mediumdose baica lin group(P>0.05).TH expression,compared with the normal group,PD group, the number of TH positive cells was significantly less,the difference was statistic ally significant(P<0.01);after drug intervention,compared with the PD group,th e low dose of baicalin positive cells(P>0.05).The number of TH positive cell in the middle and high dosage groups of meclopa and baicalin significantly increase-d(P<0.01),but the difference was not statistically significant(P>0.05)There w-as no significant difference between the baicalin group and the high dose baicalin group(P>0.05).Compared with the highdose baicalin group,the difference betw-een the baicalin group and the high dose baicalin group was not statistically sign-ificant(P>0.05).The expression of Cx43 in PD group was significantly increased by immunohistochemistry and Western Blot.The expression of Cx43 in PD grou-p was significantly lower than that in PD group(P<0.01),but Baicalin,There wa-s no significant difference between the low dose group and the PD group(P>0.05),while there was significant difference between the mid dose and high dose groups of metropisone and baicalin(P<0.01)Compared with the highdose baicalin group,there was nosignificant difference in the number of rotations of the rats in the baicalin middle dose group,the difference was no significant difference(P>0.05)Statistical significance(P>0.05).Conclusion: Baicalin can significantly impr-ove the circling behavior of PD rats,increase the activity of tyrosine hydrogenase(TH)cells and decrease the damage of PD substantia nigraneurons by 6-0HDA.The expression of Cx43 was upregulated in the expression of ASTs in the striatum of PD rats,and baicalin downregulated the expression of Cx43.
