Objective: To investigate the anti-inflammatory effects and mechanism of Mongolian Medicine Senden-4wei Tang on rabbits with knee osteoarthritis by using the modified Hulth method in healthy adult rabbits.Method:Thirty-two rabbits were modeled using the modified Hulth method:the rabbits were measured for body weight,labeled,and anesthetized with 30 g/L sodium pentobarbital in the ear vein(30 mg/kg).The rabbits were then fixed on the operating table.,knee extensions.Conventional shaving,disinfection,and drape.A longitudinal skin incision about 2.5 cm long was made on the medial side of the right knee joint of the rabbit.The medial collateral ligament was cut and the medial meniscus was completely removed and the anterior cruciate ligament was cut.When modeling,attention should be paid to protecting the articular cartilage surface from being damaged.After hemostasis,the layers are sutured and routinely sterilized.Postoperative infection tendency,will be intramuscular injection of penicillin 400,000 U / d,to prevent infection.Afterwards,they were chased three times a day.Each drive was driven for at least 30 minutes.After four weeks,the model was successful.The successful rabbits were randomly divided into three groups: the model group(8);the Senden-4 Weitang high group(8);the Senden-4 Weitang low group(8),and the remaining 8 normal homes.Rabbits were normal controls.After the treatment was completed,all groups of rabbits were sacrificed.Under aseptic conditions,the cartilage of the distal femur and proximal tibia was homogenized under low temperature conditions and centrifuged.The numbers were placed in plastic centrifuge tubes and capped separately.80 ° C freezer frozen.Finally,enzyme-linked immunosorbent assay was used to detect the proliferation of articular cartilage with PCNA,IL-1 ?,TNF-?,transforming growth factor(TGF-?)and Western blot.The effect of MMP-1,TIMP-1 protein expression in tissues.Result : 1.Compared with the normal group,the expression of MMP-1 and TIMP-1 in the articular cartilage of rabbits in the model group was significantly increased(P<0.05);compared with the model group,the MMP-1,TIMP-1 protein expression was significantly reduced(P <0.05).2.Compared with the normal group,the levels of IL-1?,PCNA,and TNF-?in cartilage tissue of the model group were significantly increased(P<0.01).Compared with the model group,the rabbit knee joints in the treatment group The levels of IL-1?,PCNA,and TNF-? in cartilage tissue were significantly down-regulated,with significant differences(P<0.05 or P<0.01).3.Compared with the normal group,the levels of TGF-? in the cartilage tissue of the knee joints of the model group were significantly increased.Compared with the model group,the levels of TGF-? in the knee cartilage tissue of the rabbits in the treatment group were significantly down-regulated,with significant differences(P<0.05 or P<0.01).Ronclusion:Through this study,it was found that changes in the levels of IL-1?,PCNA,TNF-?,TGF-?and the expression of MMP-1 and TIMP-1 proteins have an important influence on the pathogenesis of knee osteoarthritis.Decoction ofMongolian Medicine Senden-4 Decoction can inhibit or mediate the changes of IL-1?,PCNA,TNF-?,TGF-?and the expression of MMP-1 and TIMP-1 in articular cartilage of rabbits with knee osteoarthritis It has a positive effect on the treatment of knee osteoarthritis.It provides a theoretical basis for the clinical treatment of knee osteoarthritis for Mongolian Medicine Dengden-4weitang.
