Purpose: By observing the influence of Jianzhong Huoxue Xingqi Decoction on the defecation function of rats with slow transit constipation(STC),the changes of the content of substance P(SP)and vasoactive intestinal peptide(VIP)in the colon of STC rats were observed,and the Jianzhong Huoxue was explored.The therapeutic effect and mechanism of Xingqi Tang in the treatment of slow transmission constipation provide experimental basis for Jianzhong Huoxue Xingqi Decoction in the treatment of slow transmission constipation,and lays a foundation for the further clinical study of Jianzhong Huoxue Xingqi Decoction in the treatment of STC.Methods: Fifty SD rats(half males and females)weighing 160 to 180 g were selected.After 10 days of adaptive feeding without abnormalities,they were randomly divided into blank control group(n=10)and STC model group(n=40).The model group included the model control group,mosapride group,Jianzhong Huoxue Xingqi decoction low dose group,Jianzhong Huoxue Xingqi decoction high dose group.The blank group was given 0.9% normal saline 2ml/d,and normal intake of water for 21 days.The constipation model group and treatment group rats were given daily diphenoxylate 10mg/kg orally,for 21 consecutive days.And collect the number of 24 h stools after the completion of the model,measure the first black discharge time,conduct a model evaluation,determine the success of the model after the treatment.The treatment components were treated by moxabiline 1.5 mg/kg.d,by the high dose group of jianzhong hengxingqi decoction 12.69 g/kg.d,and by the low dose group 6.35 g/kg.d by gastric lavage,The number of excrement particles,wet weight and colon propulsive rate were observed in the model of slowtransmitted constipation.Results:(1)General situation: Before the model establishment,the rats in each group were moisturized,sensitive to activity,no irritability,normal diet,smooth stool,no dryness,and normal defecation.After 7 days of modeling,the rats of each model group gradually began to have dull fur,reduced fur,reduced feeding,lack of energy,decreased activity,reduced frequency of defecation,decreased stool volume,smaller stool particles,and harder stool.With Jianzhong Huoxue Xingqi Decoction and mosapride treatment started,after 7 days of treatment,the mental state of the rats in each treatment group improved compared with that before treatment.Rats' food intake increased to varying degrees,and the fur gradually became moist.The stool gradually becomes moist and the particles become larger.(2)The rate of prolapse of colonic charcoal in each group after the completion of the model: Compared with the blank group,the rate of colonic charcoal advancement in the model group was significantly decreased(P<0.05);the colon of each treatment group was compared with that of the model group.The advancing rate of charcoal was significantly increased(P<0.01).Compared with the mosapride group,the colonic charcoal pushing rate in the high-dose group of Jianzhong Huoxue Xingqi Decoction was significantly higher(P<0.05).(3)Changes in substance P and VIP levels after treatment:(1)P Substance: Compared with the control group,the average optical density of substance P in the colon of the model control group was significantly reduced,with significant statistical significance(P<0.01);after treatment The average optical density of substance P in the colon of Jianzhong Huoxue Xingqi Decoction(high and low dose)group and mosapride group was significantly increased(P<0.05).Jianzhong Huoxue Xingqi Decoction The increase of the average optical density of substance P in colon of high-dose group was more obvious than that of Mosapride group and Jianzhong Huoxue Qingqi decoction group(P<0.05).The mean optical density of substance P in colon of Jianzhong Huoxue Xingqi decoction group was not significantly different from that of mosapride group(P>0.05).(2)VIP: The level of VIP in the colon tissue of the model control group increased,compared with the blank group,the difference was significant.The content of VIP in the colon of Jianzhong Huoxue Xingqi decoction high and low dose group and mosapride group was down-regulated,and there was significant difference compared with the model control group(P<0.05).Among them,the decrease of colonic VIP in the high-dose group of Jianzhong Huoxue Xingqi decoction group was more obvious than that of mosapride group and Jianzhong Huoxue Xingqi decoction low-dose group(P<0.05).The average optical density of colonic VIP in the mosapride group was significantly lower than that of the Jianzhong Huoxue Xingqi Decoction low dose group(P<0.05).Conclusions:1.The rat model of slow transmission constipation was successfully established in this experiment.The model is highly reproducible.2.Increased VIP levels reduce intestinal motility and constipation.3.Substance P plays an important role in regulating gastrointestinal function,which can enhance the intestinal peristalsis,reduce the content of P,and may cause constipation.4.Jianzhong Huoxue Xingqi Decoction can reduce the content of VIP in the colon and increase the content of substance P in the colon,To achieve the effective treatment of STC,the mechanism of Jianzhong Huoxue Xingqi decoction for constipation may be related to the changes of enteric neurotransmitters(VIP,SP).5.This experiment also confirmed that “the deficiency of Zhongyang,blood stasis,and qi block” are the etiology and pathogenesis of STC in TCM,and “Wenzhongyiqi,Huoxuetongluo,and qi qitong” are the TCM treatment principles of TCM.According to the governing law,the “Jianzhong Huoxue Xingqi Decoction” according to this prescription is an effective prescription for the treatment of slow-transit constipation(STC).
