(1)Objective:Parkinson's disease(PD)is a common neurodegenerative disease that occurs after Alzheimer's disease.Typical neuropathological features of PD include degeneration of dopaminergic neurons located in the pars compacta of the substantia nigra that project to the striatum(nigro-striatal pathway)and depositions of cytoplasmic fibrillary inclusions(Lewy bodies)which mainly contains ?-synuclein.In recent years,endoplasmic reticulum stress is following the death of the signal path,the mitochondria pathway,a new cell apoptosis pathways,endoplasmic reticulum function can cause damage to the ERS,sustained or too strong ERS activates a series of apoptotic signals,cause apoptosis.Binding immunoglobulin heavy chain protein and Caspase12 are two key factors in ERS.Thus,the PD model was prepared by subcutaneous injection of rotenone into the rat's neck.To observe the effects of electroacupuncture "fengfu" and "taichong" on the expression of ERS related protein Bip and Caspase12 in PD model rats.(2)Methods:120 Male SD rats were randomly divided into a normal group,a sham-operation group,a model group,four EA groups(Treatment respectively for 7 days,14 days,21 days,28 days),15 rats in each group.Rats in the model group were induced by a subcutaneous injection of rotenone at the rat's nape,and rats in the sham-operation group were injected with sun-flower oil that did not include rotenone.Rats in the EA groups electroacupunctured at "Fengfu" "Taichong" point,using continuous wave,frequency of 2 Hz,the intensity of 1 mA with electroacupuncture treatment instrument of G6805-2A type,duration 20 min,once a day.Rats in the EA groups treated with electroacupuncture for 7 days,14 days,21 days,and 28 days respectively.While the normal group,sham-operation group and model group were not treated.After the treatment,behavioral score was used to evaluate the ethology changes of PD rats.Changes in the expression of ?-syn and TH positive neurons in the substantia nigra of rats were detected by immunohistochemistry and endoplasmic reticulum stress-related proteins.The changes of Bip and Caspase12 protein expression were detected by Western blot.The expression of BipmRNA and Caspase12 mRNA in the substantia nigra of rats was detected by RT-PCR.(3)Results:Behavioral results showed that rats in the model group caused by rotenone showed obvious symptoms of PD.Compared with that in model group,the behavioral score of PD rats after EA intervention was significantly improved(P<0.01);Immunohistochemical detection showed that compared with the normal group and the sham operation group,the positive expression of TH in the substantia nigra decreased significantly(P<0.01)and the positive expression of a-syn was significantly higher(P<0.01)in the model group.Compared with the model group,the positive expression of TH in the EA 7d group and EA 14 d group were increased(P<0.05),the positive expression of TH in the EA 21 d group and EA 28 d group were significantly increased(P<0.01);the positive expression of a-syn in the EA 7d group and EA 14 d group were decreased(P<0.05),the positive expression of a-syn in the EA 21 d group and EA 28 d group were significantly decreased(P<0.01).Western blot showed that compared with the normal group and sham operation group,the expression of Bip and Caspase12 in model group were significantly increased(P<0.01).Compared with the model group,the expression of Bip and Caspase12 in the EA 7d group was decreased(P<0.05),the expression of Bip and Caspase12 in the EA 14 d group?21d group and 28 d group were significantly decreased(P<0.01);RT-PCR showed that compared with the normal group and sham operation group,the expression of BipmRNA and Caspase12 mRNA in model group were increased(P<0.05).Compared with the model group,the expression of BipmRNA and Caspase12 mRNA in the EA groups decreased(P<0.05)(4)Conclusion:The rat model of PD was successfully established by subcutaneous injection of rotenone into the neck and this method is simple,economical,and has a high success rate.Electroacupuncture at "Fengfu" and "Taichong" acupoints can promote the expression of TH positive neurons and decrease the expression of ?-syn positive neurons,and ERS may be involved in the pathogenesis of PD.When high-intensity ERS induces cell apoptosis,acupuncture may prevent the damage of DA neurons in the substantia nigra by inhibiting the activity of the specific apoptotic factor Caspase12,thereby delaying the course of PD.In summary,electroacupuncture at "Fengfu" "Taichong" acupoints can protect DA neurons,which may be related to the inhibition of high-intensity ERS-induced apoptosis and decrease of the expression of specific apoptosis factor Caspase12.
