Objective:We use immunofluorescence?enzyme-linked immunosorbent assay and toluidine blue staining,and choose experimental animal C57BL/6J and homologous TRPV1-/-mice.Mice in Baihui as a starting point,observed before and after acupuncture and moxibustion intervention mice mice Baihui local skin damage of a neuropeptide,vanillin subtype-1(TRPV1)mast cells(MC),the dynamic change of material.Methods:Thirty-nine C57BL/6J and 39 C57BL/6J homologous TRPV1-/-mice were randomly divided into C57BL/6J EA group,C57BL/6J moxibustion group,C57BL/6J blank group,and TRPV1-/-EA group,TRPV1-/-moxibustion group,TRPV1-/-blank group.Three were used for immunofluorescence and 10 were used for enzyme-linked immunosorbent assay.The expression of mast cells,calcitonin gene-related peptide,substance P,serotonin,vanilloid-1,etc.was observed in each group.Experimental methods:The C57BL/6J wild type and 39 homologous TRPV1-/-mice were randomly divided into two groups,Electroacupuncture C57BL/6J group,hot moxibustion C57BL/6J group,blank C57BL/6J group and electroacupuncture TRPV1-/-group,hot moxibustion TRPV1-/-group,blank TRPV1-/-group,each group of 13 rats in each group.3 of them were used to do immunofluorescence histochemical experiments to observe the changes of mast cells,calcitonin gene-related peptide,substance P,5-HT,vanillin subtype-1,and 10 of them were used in enzyme linked immunosorbent assay to detect the content of P substance in mice skin.Result:Immunofluorescence histochemistry and enzyme-linked immunosorbent assay,C57BL/6J wild type mice:compared with the blank Baihui acupoint of C57BL/6J mice,the expression of P substance(SP),calcitonin gene related peptide(CGRP)and instantaneous receptor potential vanillin receptor(TRPV1)in the acupoints of hot moxibustion and C57BL/6J mice after C57BL/6J mice increased(P<0.05).The local TRPV1 and CGRP of the C57BL/6J acupuncture group increased significantly in the skin epidermis.C57BL/6J,SP,TRPV1 and CGRP in the Baihui group were significantly increased in the dermis and subcutaneous tissue.The proportion of "degranulation like" in the morphogenesis of 5-hydroxytryptamine(5-HT)was significantly increased by hot moxibustion and acupuncture.There was no obvious change in the expression number with the the blank group?the moxibusition group and the acupuncture.Homologous TRPV1-/-mice:the expression of SP,CGRP and TRPV1 in the acupuncture group of Baihui group increased significantly in the dermis and subcutaneous,and the expression of 5-HT was not changed,and the morphologic degranulation was changed.There was no significant difference in SP,CGRP,TRPV1 and 5-HT between TRPV1-/-hot moxibustion and Baihui group compared with blank group.Toluidine blue staining:mast cells degranulation was obvious in C57BL/6J needling Baihui group,C57BL/6J heat moxibustion Baihui group and TRPV1-/? acupuncture Baihui group.The mast cells of C57BL/6J blank Baihui group,TRPV1-/-blank Baihui group and TRPV1-/-heat moxibustion Baihui group were intact.Conclusion:TRPV1 is the initiating factor in local skin of hot moxibustion.There are similarities in the cytochemical changes of local skin between acupuncture and hot moxibustion at Baihui acupoint.
