Objective:To explore the mechanism of protease-activated receptor 2(PAR-2)and transient receptor potential vanilloid subtype 1(TRVP1)in regulating the diarrhea irritable bowel syndrome(IBS-D)process of electro-acupuncture at Zusanli,by observing the changes of m RNA and protein of PAR-2 and TRVP1 in intestinal tissue of rats with IBS-D by electro-acupuncture at Zusanli point.Methods:8 male SD rats were randomly selected as blank group,a IBS-D rat model was established by chronic restraint stress combined with senna.After successful modeling,it was randomly divided into model group and electroacupuncture group,each group has 8 rats.Electroacupuncture group took Zusanli point for electroacupuncture intervention,20 min/d,for retention continuous intervention 7d.By measuring rat weight gain,diarrhea index,stool Bristol score,to assess the colorectal distension(CRD)and abdominal wall withdrawal reflex(AWR)scores,the expression of PAR-2、TRVP1 in rat colon was determined by fluorescence quantitative PCR and Western blot.Results:1.Weight gain: Compared with the blank group after modeling,body weight gain was slow in model group and electro-acupuncture group(P<0.01);After the intervention,compared with the model group,the weight gain of electro-acupuncture group was significantly higher(P<0.01);2.The situation of diarrhea: Diarrhea index and stool Bristol score in model group were significantly higher than those in blank group and electro-acupuncture group(P<0.01);3.Visceral sensitivity evaluation: Compared with the blank group,the threshold of abdominal contraction and elevation in the CRD group decreased significantly(P<0.01),hip elevation threshold also decreased(P<0.05);Compared to the model group,abdominal contraction threshold increased significantly in electro-acupuncture group(P<0.01),and the threshold of abdominal elevation also increased(P<0.05);Compared with the blank group and the electroacupuncture group,The AWR scores in the model group increased(P<0.05),when the capacity was 1 ml,the score of the model group increased significantly(P<0.01).4.Fluorescence quantitative PCR method: The relative m RNA expression in the model group was significantly higher than that in the blank group(P<0.01),the relative expression of m RNA TRPV1 in the model group was higher than that in the blank group(P<0.05),the relative m RNA expression of PAR-2、TRPV1 in electro-acupuncture group was lower than that in model group(P<0.05)5.Western blot method: The protein expression in model group was significantly higher than that in blank group(P<0.01);Compared to the model group,the protein expression of PAR-2in electro-acupuncture group decreased significantly(P<0.01),the protein expression of TRPV1 in electro-acupuncture group also decreased(P<0.05).Conclusion:1.Electro-acupuncture at Zusanli can reduce the visceral sensitivity of IBS-D rats,to improve diarrhea in rats,is one of the effective methods to treat IBS-D.2.The expression of PAR-2 and TRPV1 in the intestinal tissue of IBS-D model rats increased,which may be related to the pathogenesis of IBS-D.3.Electro-acupuncture at Zusanli point may play a role in regulating diarrhea and visceral sensitivity of IBS-D model rats by affecting the expression of PAR-2 and TRPV1. |
