ObjectiveTo investigate the effect of NOD1 on the chemosensitivity of paclitaxel in receptor-positive breast cancer.MethodsA transient transfection model of NOD1 over-expressing MCF-7 was established by transient transfection.RT-PCR and Western blot were used to determine the mRNA and protein expression of NOD1 in transiently transfected MCF-7 cells.Firstly,Vector carried pCMV3-NOD1 was transfected into MCF-7cells transiently,then the effects of paclitaxel on cell vality,apoptosis,invasion and migration of the experimental group(MCF-7 cells over-expressing NOD1)and the control group(MCF-7 cells transfected with empty plasmids)were determined by MTT assay,flow cytometry assay,cell wound healing assay,Matrigel transwell assay.And the expression of MMP-2,Bcl-2,cleaved-caspase9 and Bax in experimental groups and control groups after paclitaxel treatment were further detected by Western blot.ResultsAfter transfection,the mRNA expression and protein level of NOD1 in MCF-7 cells that transfected with p CMV3-NOD1 were significantly increased compared with those of MCF-7 cells transfected with pCMV3-untagged and MCF-7 cells(P<0.05).The transient transfection cell model of NOD1 overexpression in MCF-7 cells was successfully established.Compared with the control groups,Paclitaxel significantly inhibited the proliferation,migration and invasion of MCF-7/NOD1 cells.Moreover,paclitaxel induced apoptosis was significantly enhanced in experimental groups.In addition,the molecular mechanism of NOD1 enhancing the therapeutic of paclitaxel was determined by Western Blot.The results showed that the effect of NOD1 on paclitaxel wassignificantly associated with expression of Bax,cleaved-caspase9,MMP-2 and Bcl-2.ConclusionNOD1 in hormone receptor positive breast cancer can increase the chemosensitivity of paclitaxel and enhance the killing effect of paclitaxel on tumor.NOD1 has no direct killing effect on tumor cells.More importantly,NOD1 enhances the efficacy of paclitaxel in breast cancer by enhance the expression of Bax protein,cleaved-caspase9 protein,inhibiting the expression of Bcl-2protein and MMP-2 protein expression.
