Objective:To observe the effect of water extract from Panax japonicus on He La cells apoptosis and its possible molecular biological mechanism,we will use human cervical cancer He La cells as the object of study for the purpose of providing the theoretical and experimental basis for the application of Panax japonicus in the treatment of cervical cancer.Methods : Firstly,the cell intervention solution containing water extract of Panax japonicas was prepared.The He La cell was routinely cultured,and it was interfered with water extract of Panax japonicas(concentration: 0.125 ~ 4 mg/ml)for 48 hours.Then,CCK-8 assay and apoptosis assay were carried out to detect the survival rate by using FCM.Meanwhile,the m RNA expression of Bax,Bcl-2 and Caspase-3 were evaluated with fluorescence quantitative real-time polymerase chain reaction(RT-PCR).Furthermore,the protein expression of Caspase-3,Bcl-2 and Bax was evaluated by western blot.Results: 1)CCK-8 assay results show that water extract of Panax japonicus plays a significant role in inhibiting the growth of He La cells with concentration of 1mg/ml ~4mg/ml in 48 hours.2)Apoptosis assay by FCM shows that the apoptosis rate of the control group is only 3.13% at the early stage,while the 1mg/ml,2mg/ml and 4mg/ml concentrations of the extracts of Panax japonicus on Hela cells are 9.11%,14.7% and 28.3%,respectively.3)Real time fluorescent quantitative PCR results show that the m RNA expression level of Bax increases,while the m RNA expression levels of Bcl-2 and Caspase-3 decrease.4)Western blot results show that water extract of Panax japonicus can down-regulate protein expression of Bcl-2 and activate apoptosis due to the overexpressed Bax and cleaved Caspase-3.Conclusion:Water extract of Panax japonicus can inhibit the growth of He La cells and induce apoptosis effectively.And the possible mechanism may be correlated with the increase of Bax,the decrease of Bcl-2 and the activated Caspase-3.
