Objective: Clinical efficacy of treatment of erhangsan was observed with treatment cervical intraepithelial neoplasia,it could provide a simple and effective methods of treatment.To further explore the mechanism of erhuangsan, hela cells were the object of study and observed its molecular mechanism for the clinical treatment of CIN and cervical cancer with a reliable basis.Prevalence of cervical intraepithelial neoplasia was discussed.Method: Clinical study: (1)onset age of illness associated with a history were analyzed,its illness trends was discussed. (2) erhuangsan was treatment of cervical intraepithelial neoplasia by analyzing before and after treatment of lesions pathological changes.Experimental study:(1) morphology of hela cells was detected by HE staining;(2) ultrastructural change of hela cells was checked by the transmission electron microscopy; (3) bcl-2 protein expression was detected by the immunohistochemical mathed; (4) survinin of mRNA expression was checked by the RT-PCR; (5) apoptosis of Hela cell was checked by the flow cytometry. Results: Clinical study: (1)from 20 to 62 years old with onset illness age , average age 38.9±8.00 years; (2) from CIN diagnosis to the frist menstrual ,it is from20 to 30 years, the average 24.0 years (3) according to relationship CIN diagnosis and times of pregnancy, the most four times pregnant, 1-2 times of pregnancy was accounted for 77%; (4) after three courses of treatment by the erhangsan ,treatment group and control group of efficient rate were: 90.0% and 66.7%. According to X2 tests and analysis,α= 0.05 for the standard,( P <0.01) it was significantly statistically significant that treatment group and control group was significant difference.Experimental study: observed by the light microscope, cell density reduced, cell shape changed: cell membrane was integrity, nuclear condensation,nuclear fragmentation:different sizes,small spherical volume,half moon shape,nucleolus disappeared, nuclear staining was purple or brownish; (2) observed by electron microscope,cell surface microvilli reduced,nuclear volume decreased,lipid droplets markedly increased, mitochondrial membrane structure was damaged and its structure unclear;(3) the expression of Bcl-2 protein decreased markedly ( P<0.01) with erhuangsan of different concentrations and times;(4) apoptosis rates of hela cells was increased with gradually mounting of time with 2.5mg/ml erhaungsan ;(5) the mRNA expression of survivin can exist in certain extent and was decreased by using erhuangsan.Conclusion:(1) erhuangsan can reduce the part inflammatory cervical reflect and enhance part immunity of the cervix;(2) erhaungsan can treat cervicalintraepithelial neoplasia, effectively prevent incidence of cervical cancer;(3) erhuangsan can induce apoptosis of hela cells in the morphological changes; (4) erhuangsan can inhibite the growth and induce apoptosis of hela cells,dependent on time and density;(5) erhaungsan induced apoptosis in hela cells, its molecular mechanism may be related to expression of Bcl-2 protein reduced and mRNA expression survinin;(6) cervical intraepithelial neoplasia may exist a trend of early age of illness.
