ObjectiveExtracellular signal regulated kinase 2(ERK2)and eye absent gene 3(EYA3)are closely related to the occurrence and development of malignant tumors.However,the mechanism by which they regulate the development and progression of retinoblastoma is not yet clear.In order to further explore their function and role in the occurrence and development of retinoblastoma,we constructed the eukaryotic expression vectors of Myc-ERK2 and EYA3 small interfering RNA(siRNA)respectively,and used the growth curve to detect their effect on the human retina Effect of WERI-Rb-1 cell growth.It is expected to provide a valuable theoretical basis for the development of retinoblastoma.MethodsThe target gene ERK2 sequence was amplified from the human breast cDNA library preserved in our laboratory by PCR and successfully inserted into Myc vector to construct Myc-ERK2 recombinant plasmid.The empty vector and the recombinant plasmids were transfected into HEK293 T cells.Western blot was used to detect the expression of ERK2.The growth curve was used to detect the effect of ERK2 on the growth of retinoblastoma(WERI-Rb-1)cells.Then,based on the cDNA sequence of EYA3,EYA3 siRNA recombinant plasmids were designed and constructed.EYA3 siRNA recombinant plasmid was transfected into HEK293 T cells,and the expression of EYA3 gene was detected by RT-PCR and Western blot respectively.The constructed EYA3 siRNA recombinant plasmid was stably transfected into WERI-Rb-1 cells,and the growth curve of EYA3 on the growth of WERI-Rb-1 cells was detected by using the growth curve.ResultsThe experiment successfully obtained Myc-ERK2 and EYA3 siRNA recombinant plasmid.Through this experiment it can be concluded that ERK2 can promote the growth of human retinoblastoma WERI-Rb-1 cells,knocking down EYA3 can inhibit the growth of human retinoblastoma WERI-Rb-1 cells.This experiment lays a foundation for further exploration of the function of ERK2 and EYA3 in retinoblastoma,and provides a new theoretical platform for the subsequent study of the occurrence and development mechanism of retinoblastoma.ConclusionsERK2 can promote the growth of WERI-Rb-1 cells,knocking down EYA3 can inhibit the growth of WERI-Rb-1 cells.This experiment lays a foundation for further exploration of the function of ERK2 and EYA3 in retinoblastoma,and provides a new theoretical basis for the subsequent study of the occurrence and development mechanism of retinoblastoma.
