?Objective?Through establishing an animal model similar to human knee ost eoarthritis,observe the effect of Modified Youguiyin on IL-1,TNF-? of serum and synovial membrane and chondrocyte apoptosis of knee osteoarthritis in ex perimental rabbits,so as to explore the mechanism of the treatment of knee os teoarthritis by Modified Youguiyin.?Methods?Forty-four normal healthy New Zealand white rabbits were selected and randomly assigned to 4 groups: 11 in the normal control group(A group),11 in the model control group(B group),11 in the positive control group(C group)and 11 in the Modified Youguiyin group(D group).The model control group,positive control group and Modified Youguiyin group were modeled by polymer plaster bandage left knee joint knee extension for 6 weeks.After successful modeling,the positive control group was given diacerein by gavage.The Modified Youguiyin group was given Modified Youguiyin by gavage.The normal control group and the model control group were given saline by gavage.After 6 weeks of intervention,four groups of animals were collected by cardiac puncture 5ml(8h fasting)for examination,all animals were sacrificed after blood collection,and the left posterior knee articular cartilage and synovial joints were removed surgically.Gross observation and light microscopy were performed on each group of articular cartilage specimens.TUNEL method was used to detect chondrocyte apoptotic index,ELISA was used to detect serum and synovial membrane IL-1,TNF-? levels,statistical analysis of experimental data,and reached a conclusion.?Results? 1.OARSI score comparison of gross observation of articular cartilage:Modified Youguiyin group was higher than the normal control group,less than the model control group(P <0.05,the difference was statistically significant).Modified Youguiyin group and the positive control group are similar(P>0.05,the difference was not statistically significant).2.Comparison of Mankin's scores of articular cartilage under light microscope: the Modified Youguiyin group was higher than the normal control group and smaller than the model control group(P<0.05,the difference was statistically significant).Modified Youguiyin group and the positive control group are similar(P>0.05,the difference was not statistically significant.3.Comparison of levels of IL-1 and TNF-? in serum and synovium: the levels of IL-1and TNF-? in the serum and synovium of the Modified Youguiyin group(D group)increased compared with the normal control group,but were lower than the model control group(P<0.05,the difference was statistically significant).The levels of IL-1and TNF-? in the serum and synovium of the Modified Youguiyin group were similar to those in the positive control group(P>0.05,the difference was not statistically significant).4.Comparison of apoptosis index of articular cartilage chondrocytes: the Modified Youguiyin group was higher than that of normal control group(P<0.05,the difference was statistically significant),but lower than the model control group(P<0.05,the difference was statistically significant).The chondrocyte apoptosis index in Modified Youguiyin group was similar to that of positive control group(P>0.05,the difference was not statistically significant).5.Correlation analysis of serum and synovial membrane IL-1,TNF-? levels and articular chondrocyte apoptosis index: There was a positive correlation between serum and synovial IL-1 levels and chondrocyte apoptotic index(all P<0.05).The correlation coefficients(r)were 0.647 and 0.819,respectively.Serum and synovial IL-1 level and chondrocyte apoptosis have a certain degree of synergistic positive change.There was a positive correlation between serum and synovial TNF-? levels and chondrocyte apoptosis index(all P<0.05).The correlation coefficient(r)were0.612 and 0.786,respectively.Serum and synovial TNF-? level and chondrocyte apoptosis have a certain degree of synergistic positive change.?Conclusions?1.Modified Youguiyin can improve the condition of knee osteoarthritis in rabbits and protect articular cartilage to a certain extent.2.Modified Youguiyin can reduce t the serum and synovial membrane IL-1 and TNF-? levels in KOA model rabbits to some extent and can inhibit chondrocyte apoptosis,which may be one of the mechanisms for its treatment of knee osteoarthritis.3.There is a positive correlation between serum and synovial membrane IL-1 and TNF-? levels and apoptosis of chondrocytes,suggesting that IL-1 and TNF-? may participate in and aggravate the apoptosis of OA articular cartilage cells,which may be one of the mechanisms of their involvement in the development of osteoarthritis.
