| Objective: The study intends to treat the treatment of AD with "Strengthing Spleen and Enriching Blood Eliminating Wind" as the treatment of the major treatment.Through clinical observation with SCORAD scores and the patients life quality index for curative effect and objective evaluation of clinical curative effect and analysis and discussion on the mechanism of action of alzheimer disease(AD),in order to improve the clinical symptoms in patients with AD,improve the quality of life for patients with scientific and effective method of treatment.Horniness cells as the experimental object,through the observation of Strengthing Spleen and Enriching Blood Eliminating Wind medicated the influence of different concentration on in vitro culture HaCat cell proliferation,an d detection of HaCat cells to understand related skin barrier function proteins in Strengthing Spleen and Enriching Blood Eliminating Wind functions of party in the experiment of HaCat cells related proteins and the effect of treatment by using the method of Strengthing Spleen and Enriching Blood Eliminating Wind functions of AD provide more reliable and scientific basis,to make better guidance for future clinical practice.Method:In clinial study,select 60 patients with age from 2 to 65 years old patients,the experimental group and positive control group 30 cases,oral Chinese medicine group,control group orally loratadine,both groups with QingPeng ointment for external use,after 4 weeks treatment,we will evaluate two groups of SCORAD scores change and serum protein levels.In addition,30 healthy groups were used as negative control,and the correlation between serum related protein content and AD patients in the healthy group was compared,and the differences in the skin barrier related proteins between AD patients and healthy people were analyzed.In the experiment part,HaCat cells were cultured in vitro,and the growth and proliferation of HaCat cells were observed under inverted microscope.Western blot was used to determine the expression level of the closely linked protein(jam-a,claudin-1,Occludin and zo-1)in HaCat cells,and to observe the effect of the treatment for proteins.Results:1.The effectiveness of the treatment group and the control group were 90% and66.66% respectively,and the difference of efficacy between the two groups was statistically significant.The score of SCORAD in both groups showed a decreasing trend during the treatment,and the treatment group was better than the control group in the fourth week after treatment,and the difference was statistically significant.No obvious untoward effect were observed in the two groups during the clinical study.After 4 weeks follow-up,the treatment group had no statistical significance in recurrence.2.Tight junction with the AD relationship analysis Based on researching into the 60 cases of AD patients and 30 cases of healthy people is closely connected to the determination of the protein in serum,Claudin-1 are negatively related with SCORAD scores,average Claudin AD group was 1.29±0.33,healt h group was 2.18±0.5,Claudin-1 is lower than normal serum levels in patients with AD.3.Growth and proliferation of HaCat cells.It can promote the proliferation of HaCat cells.With the increase of the concentration of the Concentration of TCM,the proliferation effect is more obvious.However,when the concentration of 500ug/ml is increased,it will have a certain inhibitory effect on the cells.The proliferation of HaCat cells was most obvious in the concentration of100ug/ml.4.Closely connected protein determination results.Western Blot detect the level of 4 kinds of proteins in HaCaT cells,with the increase of solution concentration,tight junction associated protein rise,which in 100ug/ml concentration when the most obvious,with concentration to rise,protein content is no longer rise.The expression of Occludin,jam-a,claudin-1 and zo-1 in the drug group was significantly higher than that in the blank group.Conclusion:TCM treatment of atopic dermatitis curative effect,compared with oral western medicine group,the patient's skin lesions and itching degree improved significantly after treatment,and no obvious adverse reaction in the treatment,in the long term,the treatment have certain advantages that is worth to promote.The results of this study showed that the closely linked protein was closely related to the onset of AD,and claudin-1 could be used as an indicator for the development of AD disease.Spleen nourishing blood functions of party can promote HaCat cells proliferation,particularly in 100ug/ml concentra tion,at the same time,the party with the increase of concentration,tight junction associated protein rose,evident when in 100ug/ml concentration,concentration to rise again,protein content is not increased. |
