Breast cancer is one of the most common tumors in women. In recent years, with in-depth study of some oncogenes and tumor suppressor genes, some progresses have been made in the diagnosis and treatment of breast cancer. However, some patients have poor prognosis because of early recurrence and distant metastasis.Gama-Glutamyl Hydrolase(GGH)is a lysosomal enzyme.Studies have shown that GGH is highly expressed in breast cancer, but its relationship with the development of breast cancer remains unclear. Here, our study identifies that the expression of GGH is obviously higher in BLBC than in other subtypes. The over-expression of GGH significantly promoted breast cancer cells migration and invasion, and caused dramatically increasing of colony formation. In addition, through tumor xenograft experiments,we found over-expression of GGH resulted in significantly enhanced tumor growth in vivo.Importantly, through clinical specimen analysis,we observed patients with high expression of GGH had poor sensitivity to chemotherapy and low survival rate, and the expression of GGH was significantly increased with the increasing of tumor grade. Therefore, our study demonstrates that GGH is critical for tumorigenicity and metastasis of BLBC cells, and may have the potency to be a molecular target in the diagnosis and treatment of breast cancer.
