| Objective: 1,To investigate the role and the potential regulatory mechanism of FOXC1 high expression in basal-like breast cancer cells.2,To explore the fluorescence imaging and functional magnetic resonance imaging characteristics in BLBC to provide method support for breast cancer diagnosis and differentiation.Materials and methods: To establish the stable cell line with FOXC1 high expression.MTT,Transwell and flow cytometry to test the effect of FOXC1 high expression on the cytobiology characteristics.Pulmonary metastasis and subcutaneous BLBC tumor-burdened nude mice models were established to observe the pulmonary metastatic lesions and tumorigenesis and the tumor volumes were measured.Then the HE staining and immunohistochemical analysis were performed.The potential regulatory pathways and the critical genes of FOXC1 high expression were analyzed by gene chip.The fluorescence imaging and functional magnetic resonance imaging were performed on the BLBC tumor-burdened nude mice models.The characteristics of fluorescence imaging and functional magnetic resonance imaging parameters were investigated by measuring the values of fluorescence,ADC,MD,MK,D and D*.Results: MDA-MB-231 cell line with stable FOXC1 high expression was established successfully.The cell proliferation ability was enhanced significantly with FOXC1 high expression group(P<0.05).The migratory and invasive ability of FOXC1 high expression group were also enhanced significantly(P<0.01),while the apoptotic cells were reduced(P<0.05).The cells of FOXC1 high expression group increased in G1 phase,while reduced in S and G2 phases(P<0.05).The metastases were observed in 3out of 5 mice in FOXC1 high expression group,whereas no metastasis was observed in the control.The average volume and weight were increased in FOXC1 high expressed tumor models(P<0.05).Ki-67 expression of the FOXC1 high expression was increased compared with the control(P<0.01),but the average MVD and IOD values had no significant differences(P<0.05).By gene chip analysis,the effect of FOXC1 high expression may be regulated by some canonical pathways and critical genes,including Aryl Hydrocarbon Receptor Signaling,p53 Signaling,Molecular Mechanisms of Cancer pathways and IL-6,SNAI2.In the subcutaneous tumors,the total fluorescence expression values in the average area were increased in FOXC1 high expression.On DWI,the mean ADC values were decreased in FOXC1 high expression group(P<0.05).On DKI and IVIM,the mean MK values increased and the mean MD,D and D* values decreased in FOXC1 high expression group,but no significant differences were found(P>0.05).Conclusion: 1,FOXC1 high expression could promote the proliferation ability,migratory ability,invasive ability of BLBC cells and inhabit the apoptosis.2,FOXC1 high expression promote the tumor growth and pulmonary metastatic ability.3,fluorescence imaging and DWI could provide some useful information for the diagnosis and differentiation for BLBC;4,FOXC1 high expression may be regulated by some canonical pathways and critical genes,including Aryl Hydrocarbon Receptor Signaling,p53 Signaling and IL-6,SNAI2. |
