| Objective:The topic of drug-induced liver injury(DILI),which is in duced by acetaminophen(APAP)in mice,observes the serum superox ide dismutase(SOD)and malondialdehyde(MDA)level for Yuan Hua Ling Gan Decoction.It explores the mechanism of the prevention abo ut the liver injury,and provides experimental data for the clinical pre vention on DILI.Methods:72 specific pathogen free male mice that originate in Kunming,are 18~22g.The mice are randomly and averagely divided into 6 groups:the normal group,the model group,the tiopronin group,the low dose group,the medium dose group,the high dose group.After 5 days of feeding to adapt the environment,the drug is given by intragastric administration once per day,for a period of 14 days.The normal group and model group are given physiological saline.the tiopronin group is given tiopronin solution.Yuan Hua Ling Gan Decoction group according to the group are given low,medium and high dose of different solution.At the end of 2 hours after gavage,the normal group is given intraperitoneal injection of normal saline,and model group,tiopronin group,Yuan Hua Ling Gan Decoction medium dose groups were given intraperitoneal injection of APAP 500mg/kg.After 4 hours,each group of mice are respectively taken blood,and liver tissue are removed and fixed with formaldehyde.Then we examine the alanine aminotransferase(ALT),aspartate aminotransferase(AST),superoxide dismutase(SOD),malondialdehyde(MDA)of mice serum.The liver was fixed in formaldehyde dicing,dehydration,paraffin embedding,sectioning and HE staining,to observe the changes of liver tissue morphology in different groups of mice.Results:1.Yuan Hua Ling Gan Decoction can reduce ALT,AST of the DILI mice’serum that induced by APAP:Compared with the normal group,the serum levels of ALT and AST in the model group are increased,and the difference between the two groups was statistically significant(P<0.05),then the model is successful.Compared with the other liver injury groups,ALT and AST levels are significantly lower in the medium dose group and the high dose group(P<0.05);There is no significant difference between the two groups of ALT and AST levels in the medium dose group and the high dose group(P>0.05).2.Yuan Hua Ling Gan Decoction can reduce MDA content,improve the serum SOD activity of the DILI mice,serum that induced by APAP:The SOD activity in serum of mice with DILI induced by APAP is decreased,and the content of MDA is increased.The levels of SOD and MDA in serum are significantly different from those in normal group,and the difference was statistically significant(P<0.05);There is differential in SOD ctivity and MDA content in the serum of the group of Yuan Hua Ling Gan Decoction the medium dose group and the high dose group,but the difference is not statistically significant(P>0.05).3.Yuan Hua Ling Gan Decoction can reduce the liver tissue injury:the injury of liver tissue for the model group is significantly changed.Compared with model group,the liver tissue injury for other liver injury groups is alleviative;the injury of liver tissue alleviated significantly in the medium dose group and the high dose group of Yuan Hua Ling Gan Decoction.Conclusion:Yuan Hua Ling Gan Decoction has a protective effect on acute DILI which is induced by APAP,and can increase the expression of SOD.the hepatoprotective effect increased with the increase of dose in a certain range.The decrease of SOD activity,and lipid peroxidation are important factors leading to acute DILI;The mechanism of liver protection of Yuan Hua Ling Gan Decoction is related to scavenging oxygen free radicals and inhibiting lipid peroxidation. |
