Objective:Observe the effects of Fu Yuan Huo Xue (FYHX)Decoction on serum bone gamma Carboxyglutamate protein (BGP), Collagen I, Transforming growth factor-β1 (TGF-β1),genetic expression of callus of fracture sides at level of mRNA and protein, during rat freature healing via paraffin section,x-ray.Methods:Closed tibial fracture of the left femur was induced in anesthetized 70 Sprague-Dawley rats using a standardized model for production of display seal fractures using a modified technique.Facture sides were fixed by percutaneous intramedullary nails(rats up to command),6 normal rats served as control group.All rats were filled with FYHX Decoction served as treatment group while normal sodium served as control group after fracture.Blood serum was determined by the expression of serum BGP, Collagen I before fracture and on the 3th,5th,7th,14th,21th day after fracture (n=6/time point).Callus of fracture were determined by the TGF-|3 and protein expression on the 3th,5th,7th,14th,21th day (n=6/time point) via reverse transcriptase polymerase chain reaction (RT-PCR) and enzyme linked immunosorbent assay (ELISA).Results:FYHX Decoction can promote the expressions of serum BGP, Collagen I, TGF-β1 of callus of fracture sides. Serum Collagen I reaches low value on the 7th day,increases gradually and peak value on the 21thday.Two groups were in accordant trend.All callus tissue TGF-β1mRNA expression increased on the 3th,14th day at the peak,eapecially on the 14th day.There is a significantly decrease on TGF-β1mRNA expression especiallyon the 7th day.The expresion of TGF-β1mRNA is in accord with its protein. The expresion of TGF-β1mRNA and its protein increased significantly on the 3th,14th,(p<0.01),21th(p<0.05),decreased significantly on the 5th,7thday in FYHX Decoction group at the time compared with the normal sodium group. FYHX Decoction can shorten the time of fracture healing.Conclusions:1.Standardized percutaneous intramedullary nails fracture of the femur in rats using a modified technique fixed have the characters of the stability,eliability,repeatbility.2.FYHX Decoction can promote the expressions of serum BGP, Collagen I, TGF-β1 of callus of fracture sides.3.FYHX Decoction can promote the fracture healing.4.TGF-β1 is a representative gene during the fracture healing.
