Aim:on the basis of Nanofat obtained from adipose tissue proposed by Tonnard,the effects of different mechanical shear on its biological characteristics were explored to find out whether there is an effective way to obtain more adipose stem cells.To provide a certain theoretical basis for clinical fat transplantation.Methods:1.Human adipose tissue acquisition: human adipose tissue was obtained with 3mm-diameter cannula according to the traditional liposuction technique of coleman.The obtained adipose tissue was centrifuged at 3000rpm/min for 3 mins and the middle part of the syringe was obtained as structural fat.2.The establishment of nano-fat model: on the ultra-clean workbench,Emulsification of the fat was achieved by shifting the fat between two 10-cc syringes connected to each other by a female-to-female Luer-Lok connector,with the set Q and t.Q=10ml/s.The nano-fat was divided into four groups(1.t=30s,2.t=60s,3.t=90s,4.t=120s),Compared with the fat content in adipose tissue after treatment,the composition proportion of adipose stem cells and other cells and the complete degree of extracellular matrix were observed between different groups.Results:it was found that the destruction of mature adipose tissue increased gradually with the increase of treatment time,and the destruction of extracellular matrix was obvious in 120 s,but not obvious below 90 s after the treatment of fat at the fixed rate of Q=10ml/s.However,the concentration of adipose stem cells increased with the increase of treatment time,and was higher at 90 s.Conclusion:high concentration of adipose stem cells can be obtained in the vicinity of 90 s with intact extracellular matrix(ECM)and homogenous emulsion,which is beneficial to injection.
