| Objective The study aims to explore the pathogenesis of bronchiolitis obliterans(BO)in rats,and evaluate the efficacy of early intervention by Chinese medicine is or not better than that of advanced treatment;to understand the effects of Huoxue Wengtong Tang on interleukin-8(IL-8)?transforming growth factor beta1(TGF-?1)expression in lung tissue of rat models;and furthermore,to provide new methods for the treatment of BO.Methods The first part,48 male SD rats were randomly divided into two groups:group A-control group(n=12)and group B-model group(n=36);and group A were divided into A3 and A7 subgroups(n=6),group B were divided into B3 ? B7 ? B3 Chinese medicine ? B7 Chinese medicine ? B7 medicine drugs and B7 control subgroups(n=6).And rats in group B were instilled with diacetyl through trachea at a dosage of 125 mg per kilogram and rats in group A were dealt with saline in the same method and same dosage;both two groups' weight were monitored and recorded.After 3 days,rats in group A3 ? B3 were executed with invasive lung function tests,blood tests and their lung tissues fixed,and B3 Chinese medicine group were given gavage with Huoxue Wengtong Tang.After 7 days;rats in group A7?B7 were also executed with the same to group A3?B3;rats in B7 Chinese medicine group?B7 medicine drug group and B7 control group were given gavage with Huoxue Wengtong Tang?prednisone and saline,respectively.And gavage was given one time per day for continuous 21 days.On the next day of last intragastric administration,pulmonary function of all these rats were tested,and then,their blood and lung tissue samples were collected.In the second part,all the tissues were then made slice,HE staining was used to observe the changes of the lung structure in each group;immunohistochemical reagent box of interleukin-8 and transforming growth factor beta1 were used to examine the contents of them,photos of each slice were got with Olympus image collecting system and analyzed with Image Pro Plusimage analysis system,and the average optical density of each sample was measured;all the data were analyzed with the software SPSS23.0 statistics system.Results(1)After 3 days,the bronchiolitis epithelial cells in the group B3 were seen to be congested,necrotic and shedding;the bronchiolitis were infiltrated with lots of inflammatory cells,a bit of collagen deposittion;and few bronchiolitis were seen luminal stenosis with proliferation.After 7 days,group B7 developed a typical BO-like changes,a large number of inflammatory cell infiltration,epithelial cell shedding,abnormal proliferation,thickening of the mucosa,and fibrosis were seen in the wall and lumen of bronchiolitis,leading to multiple bronchiolitis stenosis;(2)after 21 days treatment,the inflammation,fibrosis and occlusion degrees of bronchiolitis in group B3 chinese medicine were slighter than group B7 chinese medicine,and the difference was statistically significant(P<0.05);(3)The expression of IL-8 and TGF-?1 in group B3 and group B7 were lower than other groups,and the difference between group B3 and group B7 was statistically significant(P <0.05).Conclusions(1)Rats in group B7 was heavier than group B3 in the infiltration of tracheal inflammation,epithelial cell necrosis,fibrosis and small airway occlusion,suggesting progression process.The occurrence of BO may caused by epithelial cell damage,leading to abnormal repair,bronchiolitis fibrosis,and ultimately to small airway occlusion;It was consistent with previous pathological reports that the damage of epithelial cells,inflammatory cell infiltration,fibrosis,result in bronchiolitis occlusion;(2)On the improvement of bronchiolitis occlusion,the early intervention of Chinese medicine Huoxue Wentong Tang is better than advanced treatment;(3)Chinese medicine Huoxue Wentong Tang improves small airway occlusion maybe according to reduce IL-8,TGF-?1 expression,inhibiting inflammation and collagen deposition. |
