Objetive The aim of this study was to approach the expression of phospholipase D1 and D2 in osteosarcoma cells and their correlation with the postoperative survival time in patients with osteosarcoma.Methods 1.46 cases diagnosed as primary osteosarcoma patients were collected in the Second Affiliated Hospital of Nanhua University?Xiangya Hospital?The Second Xiangya Hospital?the Third Xiangya Hospital ? Human Provincial Tumor Hospital with pathological and follow-up information from 2008 to 2014.The two groups of pathological sections were named group A and group B.Each group has46 pathological sections of osteosarcoma and 10 pathological sections of normal precancerous tissues.Each group is in random numbers.The pathological sections of the same number in the two groups were taken from the same patient.One group of pathological sections named group A was used for the expression of phospholipase D1(PLD1),and the other group named group B for the expression of phospholipase D2(PLD2).2.Apply immunohistochemical SP method to pathologicalsections.There were several experimental procedures,including paraffin sections dewaxed to water,antigen retrieval,blocking endogenous peroxidase,and BSA blocking.Two groups were added PLD1 and PLD2 antibody,DAB color,nuclear restain,dehydration and blocking individually.Finally scan images and capture images of pathological sections Then capture images to make data analysis according to experimental requirements.3.Use the statistical software SPSS 20 to make statistical analysis of this experimental data and make a overall data statistics.T-test was used to analyze the averages between the two groups.And spearman correlatiom coefficient was used for the expression correlation of PLD1 and PLD2.The survival curve was analyzed by Kaplan-Meier method.Values of P<0.05 were considered to indicate statistical significance.Results 1.All the 46 patients in the experiment were followed up for an average of 21.3 months(3-79).Of these patients,25 had recurrence and metastasis of osteosarcoma and 22 died.Only 15 patients survived without tumor.2.Under 200 fold microscope observation,the expression degree of PLD1 and PLD2 in osteosarcoma can be divided into 4 grades:strong positive,moderate positive,weak positive and negative.There is no significant difference in the expression level in the normal precancerous tissues.3.There were one couple of pathological sections in group A and group B which were found no result inimmunohistochemistry score.Immunohistochemical score showed that the average scores of PLD1 and PLD2 in osteosarcoma were higher than those in normal precancerous tissues.And the difference was statistically significant(p<0.05).4.The expression of PLD1 and PLD2 was positively correlated in osteosarcoma.5.Kaplan-Meier survival analysis indicated that the expression levels of PLD1 and PLD2 were negatively correlated with postoperative survival time.Conclusion The level of PLD1 and PLD2 expression in osteosarcoma increases and that expression level are negatively correlated with postoperative survival time.
