Objective:To study the expressions of membrane type-1 matrix metalloproteinase(MT1-MMP)and phospholipase D2(PLD2)in different breast tissues,and the relationship between the expression of the both and the invasion of breast cancer and the correlation between the both in the invasion and development of breast cancer.Methods:245 paraffin samples were collected.Among them,138cases(56.33%)are invasive ductal carcinoma(IDC),37 cases(15.10%)are ductal carcinoma in situ with microinvasion(DCIS-MI),46 cases(18.78%)are ductal carcinoma in situ(DCIS),24 cases(9.80%)are normal breast tissue.The expressions of PLD2 and MT1-MMP in different breast tissues were detected by immunohistochemistry,and analyzed the relationship between the expressions of the both and the invasion of breast cancer and the correlation between the both in the invasion and development of breast cancer.SPSS19.0 software system was used for statistical analysis of data,chi-square test was used for counting data analysis,Fisher exact probability method was used for comparison between two indexes,and Spearman correlation test was used for correlation analysis.P<0.05 was considered to be statistically significant.Results:1.The positive expression of MT1-MMP in normal breast tissue is 3,and the positive rate is 12.50%.The positive expression in DCIS is 16,and the positive rate is 34.78%.The positive expression in DCIS-MI is 16,and the positive rate is 43.24%.The positive expression in IDC is 81,and the positive rate is 58.70%.The positive rate of MT1-MMP in normal breast tissue,DCIS,DCIS–MI and IDC increases by degress,the difference of positive rate between the four groups is statistically significant.(?~2=21.983,P=0.000).The positive rates of MT1-MMP in DCIS,DCIS-MI and IDC are higher than that of normal breast tissue,and the differences are statistically significant(P<0.05).The positive rate of MT1-MMP in IDC is higher than that in DCIS,and the difference is statistically significant(?~2=7.915,P=0.005).The positive rate of MT1-MMP in DCIS-MI is higher than that in DCIS,but the difference is not statistically significant(?2=0.620,P=0.431).The positive rate of MT1-MMP in IDC is higher than that in DCIS-MI,but the difference is not statistically significant(?2=2.820,P=0.093).2.The positive expression of PLD2 in normal breast tissue is 4,and the positive rate is 16.67%.The positive expression in DCIS is 28,and the positive rate is 60.87%.The positive expression in DCIS-MI is 21,and the positive rate is 56.76%.The positive expression in IDC is 108,and the positive rate is 78.26%.The difference of positive rate between the four groups is statistically significan t(?~2=37.064,P=0.000).The positive rate of PLD2 in normal breast tissue is significantly lower than that in DCIS,DCIS-MI and IDC,and the differences are statistically significant(P<0.05).The positive rate of PLD2 in IDC is higher than that of DCIS group and DCIS-MI group,and the differences are statistically significant(P<0.05).The positive rate of PLD2 in DCIS-MI is lower than that of DCIS group,but the difference is not statistically significant(?~2=0.143,P=0.705).3.The expression of MT1-MMP in IDC is associated with lymph node involvement,pTNM stage,intravascular tumor,ER,HER2,Ki-67and molecular classification(P<0.05).There is no correlation with age,tumor size,distant metastasis,WHO classification and PR(P>0.05).4.The expression of PLD2 in IDC is associated with tumor size,lymph node involvement,pTNM stage,intravascular tumor and HER2(P<0.05).There is no correlation with age,distant metastasis,WHO classification,ER,PR,Ki-67 and molecular classification(P?0.05).5.In breast cancer tissue,MT1-MMPand PLD2 are located in the same location:the cytoplasm and the cell membrane.By the Spearman correlation analysis method,the expressions of MT1-MMP and PLD2 in breast cancer tissue are positive correlation(r=0.616,P=0.000).Conclusion:1.The positive rates of MT1-MMP and PLD2 in breast cancer tissue are significantly higher than that of normal breast tissue.The positive rates of the both in IDC group are significantly higher than that of DCIS group.It is suggested that MT1-MMP and PLD2 may play a role in the invasive development of breast cancer.2.In IDC,the positive expression rate of MT1-MMP is higher with more lymph nodes involvement,later pTNM stage,ER(-),HER2(+),high expression of ki-67 and positive vascular tumor.The positive rates of MT1-MMP of HER2 overexpression type and Luminal B are higher than that of Luminal A.It is suggested that MT1-MMP is overexpression in high invasive breast cancer.3.In IDC,the positive expression rate of PLD2 is higher with larger tumor,more lymph nodes involvement,later pTNM stage,positive vascular tumor and HER2(+).It is suggested that PLD2 is overexpression in high invasive breast cancer.4.MT1-MMP and PLD2 are located in the same location of tumor tissue:the cytoplasm and the cell membrane.and the expressions of the both in breast cancer tissue are positive correlation.It is suggested that there may be some synergistic effect between the both in the invasion and development of breast cancer.
