Objective:The study aimed to learn the expression of PLD1 in pancreatic cancer.Meanwhile,the correlation between PLD1 expression and the parameters of the patients was also investigated.Finally,we also examined the possible mechanism whereby PLD1 facilitates pancreatic cancer progression.Materials and Methods:1.To examine the expression and the biological significance of PLD1 in pancreatic cancer.To this end,PLD1 expression was detected in the clinical samples of pancreatic cancer.Subsequently,the correlation between PLD1 and the clinicopathological parameters of the patients were also analyzed.Finally,PLD1 expression in pancreatic cancer cell lines was also examined using western blot.2.To examine the expression and the biological significance of Spl in pancreatic cancer.To this end,Spl expression was detected in the clinical samples of pancreatic cancer.Subsequently,the correlation between Spl and the clinicopathological parameters of the patients was also analyzed.Finally,Spl expression in pancreatic cancer cell lines was examined using western blot.3.To explore the correlation between PLD1 and Spl as well as their combinational effect on the progression of pancreatic cancer.Moreover,the correlation between PLD1 and Spl was also confirmed in cell lines of pancreatic cancer.Results:1.Based on the immunohistochemical data,we found that PLD1 expression was elevated,and it is responsible for tumor vascular invasion in pancreatic cancer.The survival analysis indicated that elevated PLD1 confers a poor prognosis for patients with pancreatic cancer,which functions as an independent prognostic factor for the disease.Finally,we showed by western blot that PLD1 was also upregulated in pancreatic cancer lines.2.The immunohistochemical data showed that Spl was elevated and responsible for tumor vascular invasion in pancreatic cancer.Kaplan survival analysis indicated that elevated Spl confers a poor prognosis for the patients,which also functions as an independent prognostic factor for the disease.Finally,we showed that Sp1 was also upregulated in pancreatic cancer.3.The correlation indicated that elevated PLD1 expression was positively correlated with Sp1 in pancreatic cancer tissues.Moreover,Kaplan-Meier analysis in combination with COX regression analysis showed that the co-high expression of PLD1 and Sp1 confers the worst prognosis,and it functions as an independent prognostic factor for the lethal disease.Meanwhile,we also showed that the positive correlation could also observed in the cell lines of pancreatic cancer.Conclusion:Both PLD1 and Sp1 were pro-tumoral in pancreatic cancer.Moreover,they could synergistically promote pancreatic cancer progression.These data indicated that PLD1 together with Spl was a potential therapeutic target for the lethal disease.
