Objective To investigate clinical significance of kidney-type glutaminase(GLS1)in hepatocellular carcinoma,and its effects on proliferation,apoptosis and other biological functions of HepG2 cell line.Methods The expression of GLS1 in 66 hepatocellular carcinoma simples were evaluated by immunohistochemistry,and analyze the relationship between the expression of GLS1 and the clinicopathological index of these patients.And cell proliferation,cell cloning experimens,wound healing assay and apoptosis experiment were performed to determine the effect of GLS1 on biological function of HepG2.Results The expression of GLS1 was closely related to TNM staging(?~2=7.273,P<0.05).The overall survival(OS)and disease-free survival(DFS)in the GLS1-high expression group was lower than that in the GLS1-low expression group(P<0.05).Inhibiting the activity of GLS1 can inhibit the proliferation,clone formation assay and migration of hepatoma cells,and can promote the apoptosis of hepatoma cells.Conclusion The high expression of GLS1 is associated with clinicopathological features of patients with hepatocellular carcinoma,and can affect the prognosis of patients. |
